补肾活血中药复方对去卵巢大鼠骨密度和骨组织神经肽Y的影响 被引量:7

Effects of Nourishing Kidney and Activating Blood Recipe on bone mineral density and NPY in plasma and bone tissue in ovariectomized rats

摘要 目的探讨补肾活血中药复方对去卵巢大鼠骨密度及血浆和骨组织神经肽Y(NPY)的影响及对防治骨质疏松的作用机制。方法选择6月龄SD雌性大鼠32只,随机分为假手术组、模型组、补肾活血中药复方低剂量组和高剂量组。模型组、复方低剂量组与高剂量组均行双侧去卵巢手术,手术1 w后中药复方低、高剂量组用灌胃法1.16 g/100 g和10.44 g/100 g给服中药,模型组灌服生理盐水,12w后观察各组大鼠骨密度的改变,采用实时荧光定量(RT-PCR)检测骨组织NPY m RNA的表达美联免疫吸附(ELISA)法检测各组大鼠血浆和骨组织NPY含量,Western印迹检测骨组织NPY的表达。结果与假手术组比较,模型组骨密度明显降低(P<0.01),骨组织中NPY蛋白和m RNA的表达量明显升高(P<0.01),血清和骨组织中NPY的含量也明显升高(P<0.01)。中药复方低剂量和高剂量组骨密度明显高于模型组(P<0.01),血浆和骨组织中NPY蛋白和m RNA的表达量明显降低(P<0.01或0.05),但仍高于假手术组(P<0.01)。结论补肾活血中药复方能够调节血浆和骨组织的NPY,这可能是其抗骨质疏松的作用机理之一。 Objective To investigate the effects of Nourishing Kidney and Activating Blood Recipe ( NKABR) on bone mineral den-sity and the levels of neuropeptide Y ( NPY) of plasma and bone tissue in ovariectomized rats.Methods 32 SD rats were randomly divided into sham-operated,model,NKABR low-dose and high-dose groups.After ovariectomy 1 w,NKABR groups were given intragastric administra-tion with different doses of NKABR,model group was given normal physiological saline.After 12 w,the changes of bone mineral density was observed,the levels of NPY mRNA or protein in plasma and bone tissue were measured by real-time RT-PCR,ELISA or Western blot.Re-sults Compared with that of sham-operated group,BMD of model group was decreased significantly (P<0.01),the levels of NPY mRNA and protein of serum or bone tissue were increased significantly(P<0.01).The bone mineral density of NKABR low dose and high dose group were significantly higher than those of model group( P<0.01) ,the levels of NPY mRNA and protein of plasma or bone tissue were significant-ly lower than those of model group (P<0.01 or 0.05),but still higher than those of sham-operated group(P<0.01).Conclusions NKABR could regulate level of NPY,which might be one of anti-osteoporosis mechanisms.
