天花粉蛋白通过下调p-ERK及CyclinD1表达抑制PC3细胞增殖 被引量:4

摘要 目的探讨天花粉蛋白(TCS)体外对前列腺癌细胞(PC3)生长的抑制作用及可能机制。方法采用噻唑蓝(MTT)检测TCS对PC3细胞的抑制作用。流式细胞术(FCM)检测TCS对PC3细胞周期的影响,Western印迹检测TCS对ERK、p-ERK及细胞周期调节蛋白Cyclin D1表达的影响。结果 MTT结果显示TCS能有效抑制PC3细胞的生长,具有时间及剂量依赖性。流式细胞检测发现TCS能够将PC3细胞阻滞于G1期,Western印迹检测TCS能抑制ERK磷酸化及细胞周期调节蛋白Cyclin D1的表达。结论 TCS对PC3细胞的增殖具有明显的抑制作用,其作用机制可能与抑制丝裂原活化蛋白激酶(MAPK)细胞增殖信号通路及降低Cyclin D1表达,从而诱导细胞发生G1期阻滞有关。 Objective To explore the inhibition effect of Trichosanthin( TCS) on prostate cancer PC3 cells and investigate the possible mechanism.Methods MTT assay was used to detect the proliferation inhibition effect of TCS on PC3 cells,cell cycle was analyzed by flow cytometry.Expressions of p-ERK and Cyclin D1 protein were detected by Western blot.Results TCS could inhibit the growth of PC3 cell in a time and dose dependent manner.FCM detection showed that TCS could arrest PC3 cells in G1 Phase.Western blot found that the protein expressions of p-ERK and Cyclin D1 were decreased after treated with TCS.Conclusions TCS could inhibit the proliferation of PC3 cells by down-regulating the mitogen activated protein kinase pathway and inducing the G1 phase arrest.
