量子点多重染色在卵巢癌体外成像的应用

摘要 目的利用量子点(QDs)免疫荧光技术对卵巢癌SKOV3细胞上叶酸受体(FR)及人附睾蛋白(HE)4进行定位,初步揭示QDs多重染色在卵巢癌细胞体外成像的应用价值。方法采用活泼酯法,构建不同发射波长的QDs-链霉亲和素(SA)复合物,分别与生物素化-叶酸(FA)及生物素化-抗HE4抗体结合,形成特异性荧光探针,同时靶向结合SKOV3细胞,实现细胞双重染色。结果构建的QD-FA及QD-抗HE4抗体靶向探针具有极强的特异性,量子点多重染色的荧光强度与单重染色相比差异无统计学意义(P>0.05)。量子点具有极强的光学稳定性。结论量子点多重染色在卵巢癌细胞体外成像中具有一定的应用价值,可为卵巢癌早期检测提供一种新思路。 Objective To detect the folate receptor( FR) and human epididymis protein 4( HE4) receptor localization in ovarian cancer SKOV3 cells simultaneously by quantum dots( QDs) immunofluorescence technique,and validate the application value of multiplexed QDs staining. Methods The streptavidin( SA)-QDs compounds with different emission wavelength was established by active ester method.These compounds could be coupled with biotinylated folate and biotinylated anti-HE4 antibody,forming specific fluorescent probes to target bind SKOV3 cells simultaneously,to realize cells double staining. Results( 1) The QD-FA and QD-HE4 target probes had strong specificity.( 2) There was no significant difference in fluorescence intensity between QDs multiple staining and single staining.( 3) QDs had strong optical stability. Conclusions Multiplexed QDs staining has its application value in ovarian cancer in vitro imaging and could be a new thinking in ovarian cancer early detection.
