二甲双胍对大鼠肾小球系膜细胞氧化应激的影响 被引量:3

摘要 目的探讨不同浓度二甲双胍对高糖环境下大鼠肾小球系膜细胞氧化应激的影响及其对糖尿病肾病(DN)的保护机制。方法将大鼠肾小球系膜细胞分别培养在正常对照组(NC组)、高糖(HG组)及高糖+不同浓度二甲双胍组。48 h后,比色法测定细胞上清液超氧化物歧化酶(SOD)、丙二醛(MDA)含量,RT-PCR方法测定细胞p22phox和p47phox mRNA表达情况。结果与NC组比较,HG组MCs p22phox和p47phox mRNA表达明显增加,上清液SOD活力明显降低,MDA含量明显增高(P<0.05)。二甲双胍干预高糖组MCs p22phox和p47phox mRNA表达明显低于HG组(P<0.05),且二甲双胍浓度越高,MCs p22phox和p47phox mRNA表达越低。结论二甲双胍可降低高糖环境下大鼠肾小球系膜细胞3-磷酸甘油醛脱氢酶(GADPH)氧化酶表达水平,从而起到减轻氧化应激的作用,该作用在二甲双胍治疗糖尿病时表现为对肾脏的保护作用。
