摘要
目的研究洛伐他汀对心肌细胞中转化生长因子(TGF)-β1和白细胞介素(IL)-6表达水平的影响。方法大鼠心肌细胞株H9c2(2-1)进行体外培养过程中,以100 nmol/L洛伐他汀处理72 h,采用蛋白免疫印迹杂交检测TGF-β1和IL-6蛋白表达水平,然后进行量化比较分析。结果洛伐他汀处理导致心肌细胞TGF-β1和IL-6蛋白表达水平分别下降到对照水平的(69.70±20.89)%和(56.26±25.04)%,两组内处理组和对照组的差异均具有统计学意义(TGF-β1组:P=0.013;IL-6组:P=0.008)。结论洛伐他汀在心肌细胞中抑制TGF-β1和IL-6的表达,该效应可能是洛伐他汀抗炎机制的关键因素之一。
Objective To investigate the regulation of lovastatin on transforming growth factor( TGF)-β1 and interleukin( IL)-6 in cardiac myocytes. Methods Rat myocardium H9c2( 2-1) cells in culture were treated with 100 nmol / L lovastatin for 72 h. The protein levels of TGF-β1 and IL-6 were detected through immunoblotting and then quantitatively comparing. Results After treatment with lovastatin,the average protein levels of myocardial cellular TGF-β1 and IL-6 were decreased to( 69. 70±20. 89) % and( 56. 26 ±25. 04) % respectively with statistical significances for both TGF-β1( P = 0. 013) and IL-6( P = 0. 008). Conclusions Lovastatin inhibits protein levels of both TGF-β1 and IL-6 in cardiac myocytes,which might constitute a key factor in the anti-inflammation mechanisms of lovastatin.
