摘要
目的研究补阳还五汤(BYHWD)对阿尔茨海默病(AD)小鼠NF-κBp65蛋白表达的影响。方法小鼠随机分成对照组、模型组、治疗组(吡拉西坦0.62 g·kg-1·d-1)、BYHWD高剂量组(37.06 g·kg-1·d-1)、BYHWD中剂量组(18.53 g·kg-1·d-1)、BYHWD低剂量组(9.26 g·kg-1·d-1),连续治疗28 d后取材。显微镜观察小鼠海马镜下形态结构。免疫组化、实时定量PCR及其Wester印迹检测AD小鼠NF-κBp65蛋白的表达。结果与对照组比较,模型组小鼠神经元数量减少、发生变性,NF-κBp65蛋白水平明显升高(P<0.05);与模型组比较,BYHWD组海马神经元细胞形态改善明显,NF-κBp65蛋白水平明显降低。结论 BYHWD能维持AD小鼠神经元的正常形态结构,降低NF-κBp65蛋白水平。
Objective To explore the effect of Buyang Huanwu decoction( BYHWD) on the NF-κBp65 protein in Alzheimer’s disease( AD) mouse. Methods The mice were randomly divided into control,model,treatment( piracetam 0. 62 g · kg- 1·d- 1),BYHWD high-dose( 37. 06 g·kg- 1·d- 1),BYHWD middle-dose( 18. 53 g·kg- 1·d- 1),BYHWD low-dose( 9. 26 g·kg- 1·d- 1) groups. The mice were killed after 28 days continuous treating. After the treatment,all animals were sacrificed,morphology of hippocampal was observed.Immunohistochemical,real time quantitative PCR,Western blot were used to detect the content of NF-κBp65 protein in the hippocampal. Results Compared with those of control group,the mice in model group lost normal morphological structure of nerve cells and the number of nerve cells was significantly decreased,NF-κBp65 protein was significantly increased( P < 0. 05). Compared with that of model group,the brain maintained normal morphological structure of nerve cells,NF-κBp65 protein was significantly decreased( P < 0. 05) in BYHWD group.Conclusions BYHWD could maintain normal morphological structure of nerve cells of AD mice,decrease NF-κBp65 protein responses.
