摘要
目的研究附子和附子干姜配伍对心衰大鼠血流动力学和环磷酸腺苷(c AMP)-蛋白激酶A(PKA)信号转导通路的影响。方法采用盐酸普罗帕酮注射液静脉注射法建立心力衰竭动物模型,实验分为空白组、模型组、附子高、低剂量组(5、2.5 g/kg)、附子∶干姜(1∶1)高、低剂量组(10、5 g/kg),记录左心室内压(LVSP)、左心室内压最大上升/下降速率(±LVdp/dtmax);采用酶联免疫吸附法检测心脏组织中c AMP、PKA的含量。结果与模型组比较,给药组均可以升高LVSP和±LVdp/dtmax,其中附子∶干姜(1∶1)高剂量组效果最显著;与模型组比较,各组给药后心脏组织中c AMP和PKA含量均增加,其中附子∶干姜(1∶1)高剂量组最显著。结论附子干姜配伍有明显的抗心衰作用,可升高心肌组织c AMP、PKA含量,这可能是其作用机制之一。
Objective To observe the effects of aconite and compatibility of aconite dried ginger on hemodynamic parameters and cylic adenosine monophsphate( c AMP)-protein kinase A( PKA) signal transduction pathways in rat of chronic heart failure. Methods The acute heart failure rat animal model was established by propafenone hydrochloride injection venous injection method. The SD rats were randomly divided into normal,model,aconite( 5,2. 5 g / kg),compatibility of aconite dried ginger( 1∶ 1) group( 10,5 g / kg). The hemodynamic parameters left ventricular systolic pressure( LVSP) and maximal derivative of left ventricular pressure( ± LVdp / dtmax) were measured.Through ELISA kit,the contents of rat heart tissue c AMP and PKA were detected. Results The levels of LVSP and ± LVdp / dtmax were increased significantly in all groups,the contents of c AMP and PKA were significantly increased. Compatibility of aconite dried ginger had more significant effect in the high dose group. Conclusions Compatibility of FG has curative effects to acute heart failure,the mechanism may be related to increasing the contents of c AMP and PKA.
