摘要
目的探讨天麻多糖对H22荷瘤小鼠细胞周期及caspase蛋白活性的影响。方法右侧腋窝接种H22肿瘤细胞制造荷瘤小鼠模型。实验设模型组、高剂量天麻多糖组、中剂量天麻多糖组、低剂量天麻多糖组及环磷酰胺(CTX)组。造模后给予天麻多糖或环磷酰胺治疗。实验结束处死小鼠,比较各组瘤重、抑瘤率、caspase-3,8,9及细胞周期分布状况。结果天麻多糖可明显减轻瘤重,高剂量天麻多糖抑瘤率可达44.7%;与模型组比较,天麻多糖中、高剂量组可显著升高caspase-3,8,9水平及G0/G1期细胞百分数,降低G2/M期细胞百分数。结论天麻多糖对H22荷瘤小鼠具有显著的抑瘤作用,其作用机制可能与影响细胞周期分布,抑制细胞增殖并激活Caspase系统诱导肿瘤细胞凋亡有关。
Ob jetc ive To observe the effects of Gastrodia Elata B 1 Polysaccharide on cell cycle and caspase-3,8,9 activities in H22 tumor bearing mice.Methods Sixty mice were randomly divided into model,CTX,Gastrodia Elata B1 polysaccharide high,middle,low groups.Two weeks later ,tumor weight of mice,tumor inhibition rate,caspase 3,8,9 activities and cell cycle were inspected respectively .Re-sults Gastrodia Elata B1 Polysaccharide decreased tumor weight significantly , tumor inhibition rate in Gastrodia Elata B 1 polysaccharide high group got 44.7%.Compared with those of model group ,polysaccharide in middle and high groups increased caspase-3,8,9 activities and percent of G0/G1 phase cells,decreased percent of G2/M phase cells.Conclusions Gastrodia Elata B1 polysaccharide has obvious anti-tumor effect on H22 tumor bearing mice .The mechanism might be related to regulating the cell cycle distribution ,inhibiting the cell prolifera-tion and activating the caspase system to induce apoptosis .
