加味孔圣枕中丹对局灶性脑缺血模型大鼠神经细胞凋亡的影响 被引量:2

摘要 目的研究加味孔圣枕中丹对局灶性脑缺血模型大鼠神经细胞凋亡及凋亡相关蛋白Bcl-2、P53表达的影响。方法线栓法建立局灶性脑缺血大鼠模型,随机分为模型组、银杏叶组及加味孔圣枕中丹组,另设假手术组(n=10)。灌胃给药,连续14 d。观察大鼠神经功能症状评分,荧光TUNEL法检测神经细胞凋亡,免疫组织化学法测定Bcl-2、P53表达。结果与模型组比较,加味孔圣枕中丹能显著降低局灶性脑缺血大鼠Zea Longa和网屏实验评分,降低神经细胞凋亡率和P53蛋白表达,增强Bcl-2蛋白表达(P均<0.05)。结论加味孔圣枕中丹能改善脑缺血引起的神经功能损伤,其机制可能与调控凋亡相关蛋白表达、抑制脑缺血引起的神经细胞凋亡相关。 Objective To study the impacts of modified Kongsheng Zhenzhong pills on apoptosis,Bcl-2 and P53 expressions of neural cells in focal cerebral ischemia rats.Methods The model of middle cerebral artery occlusion( MCAO) was established by suture method.30 MCAO rats were randomly divided into model,ginkgo leaf and modified Kongsheng Zhenzhong pills groups.The neurological symptom scores were observed for the analysis of neural behavior.Fluorescent TUNEL assay was used to detect neural cells apoptosis.Immunohistochemistry was employed to determine the expressions of apoptosis-related protein Bcl-2 and P53.Results Compared with those of model group,the scores of Zea Longa and screen test,the apoptosis rate of neural cells and the expression of P53 were reduced,while the expression of Bcl-2 was increased significantly in modified Kongsheng Zhenzhong pills group( P<0.05).Conclusions Modified Kongsheng Zhenzhong pills could improve the nerve function injury of MCAO rats by modulating the expression of apoptosis-related protein and inhibiting the neuronal apoptosis.
