慢病毒介导的CDK2-shRNA促进黑色素瘤细胞A375凋亡 被引量:4

摘要 目的探讨慢病毒介导的细胞周期蛋白依赖性激酶(CDK)2-shRNA对人黑色素瘤细胞A375凋亡的影响。方法依据CDK2基因序列,设计3条靶向干扰CDK2的序列,构建CDK2-shRNA慢病毒载体,质粒转染人胚肾细胞株(HEK293)细胞进行慢病毒包装,滴度测定包装的重组p GMLV-CDK2-shRNA慢病毒,免疫印迹检测对CDK2的干扰效率及对细胞周期蛋白(cyclin)E、E2F1、RB蛋白表达的影响,四甲基偶氮唑蓝比色(MTT)检测细胞增殖活力,Annexin V-FITC/PI染色流式细胞仪检测A375凋亡的发生。结果成功获得包装的重组p GMLV-CDK2-shRNA慢病毒,病毒滴度为5×108TU/ml。重组的p GMLV-CDK2-shRNA慢病毒能有效下调CDK2、cyclin E、E2F1蛋白表达,抑制A375细胞增殖和触发凋亡。结论基于慢病毒介导的CDK2-shRNA能够促进黑色素瘤细胞A375凋亡发生,为黑色素瘤的基因治疗提供理论支撑。
