Toll样受体-9在细菌DNA诱导小鼠心肌炎症反应中的作用

摘要 目的探讨Toll样受体-9(TLR-9)及其致病性配体(即细菌DNA)在脓毒症小鼠心肌炎症反应中心功能不全的作用。方法使用人工合成的细菌DNA(Cp G-ODN),分别作用于TLR-9组及TLR-9缺陷(TLR9-D)的小鼠,测定6 h时间点炎症标志物:肿瘤坏死因子(TNF)-α、白细胞介素(IL)-1β及IL-6、血清脑钠肽(BNP)水平及心动超声的心功能结果。结果在野生型小鼠,Cp G-ODN导致心脏发生严重炎症反应,表现为TNF-α、IL-1β,IL-6及BNP的水平升高及心动超声示心脏收缩机舒张功能明显降低(P<0.05);而在TLR9-D小鼠这种炎症反应却没有出现。结论细菌DNA通过TLR-9的介导,导致心肌炎性反应及细胞因子的生产并且产生心功能不全,TLR-9与心功能不全呈正相关。
