Dickkopf-1对SD大鼠血管平滑肌细胞增殖、迁移及表型转化的影响

摘要 目的观察Dickkopf(DKK)-1对SD大鼠血管平滑肌细胞(VSMC)增殖、迁移及表型转化的影响。方法取大鼠主动脉贴块培养VSMC,不同浓度外源DKK-1分别作用于VSMC,然后采用cck8和transwell试验分别检测DKK-1对大鼠VSMC增殖及迁移的影响。RT-PCR和Western印迹分别用于检测VSMC去分化表型标志基因骨桥蛋白(OPN)mRNA和蛋白表达。结果外源性DKK-1干预后,随着作用时间延长,DKK-1浓度增大,细胞增殖率降低,迁移能力减低;还可使VSMC发生表型转化,而在这个过程中OPN mRNA及蛋白表达水平明显降低。结论外源DKK-1能抑制大鼠VSMC增殖、迁移能力,并具有剂量和时间依赖性,此外,还可使VSMC从合成型向收缩型转化。
