摘要
目的探讨中药塌渍疗法对膝骨关节炎(KOA)兔模型骨节软骨基质金属蛋白酶(MMP)-3、MMP-13、骨形态发生蛋白(BMP)-2、cylinD1、c-myc mRNA表达的影响。方法选择石膏管型固定法制作新西兰兔KOA模型,随机分为空白组、模型组、治疗组(扶他林组、42℃中药塌渍治疗组、37℃中药塌渍治疗组、32℃中药塌渍治疗组),给药4 w后处死动物解剖右膝关节软骨,切片,HE染色后行病理学Mankin评分,采用RT-PCR检测MMP-3、MMP-13、BMP-2、cylinD1、c-myc mRNA的表达量。结果病理结果显示空白组关节软骨表面光滑,未见裂痕,有光泽,模型组关节软骨表面不平,色泽暗淡,有细小骨赘生成等,治疗组关节软骨表面均好于模型组; Mankin评分模型组最高,空白组最低,治疗组评分均显著低于模型组高于空白组(均P<0. 05),治疗组从高到低依次是32℃中药塌渍治疗组、扶他林组、37℃中药塌渍治疗组、42℃中药塌渍治疗组。PCR检测结果显示模型组MMP-3、MMP-13、BMP-2、cylinD1、c-myc mRNA的表达水平均高于空白组,各治疗组表达量均显著低于模型组(均P<0. 05),治疗组从高到低依次是32℃中药塌渍治疗组、扶他林组、37℃中药塌渍治疗组、42℃中药塌渍治疗组。结论中药塌渍疗法能够降低KOA模型兔软骨MMP-3、MMP-13、BMP-2、cylinD1、c-myc mRNA的表达,改善关节软骨病理。
Objective To explore Chinese medicine collapse stains for knee osteoarthritis (KOA) rabbit model of condyle car-tilage MMP-3,MMP-13,BMP-2,cylinD1,c-myc mRNA expression.Methods KOA model of New Zealand rabbits was made by plastertube fixation and randomly divided into blank,model and treatment groups including diclofenac emulsion,42 ℃ treatment,37 ℃ treat-ment and 32 ℃ treatment groups. After 4 weeks of treatment,animals were killed and anatomy of the right knee joint cartilage,slicewas obtained to detect Mankin ’s score by HE stain. The expressions of MMP-3,MMP-13,BMP-2,cylinD1 and c-myc mRNA were de-tected by RT-PCR.Results Pathological results showed that articular cartilage surface was smooth and glossy,there was no crack inblank group. Articular cartilage surface was not smooth,colour and lustre was dim and small osteophytes in model group. Articular car-tilage surface was better than that of model group in treatment groups. Mankin’ s score was highest model group and lowest in blankgroup,Mankin’s scores of treatment groups were lower than those of model group and higher than those of blank group. From high tolow,Mankin’s score of treatment groups was 32℃ collapse stains treatment group,Diclofenac emulsion group,37℃ collapse stainstreatment group,42℃ collapse stains treatment group. PCR detection results showed that MMP-3,MMP-13,BMP-2,cylinD1,c-mycmRNA expression level of model group were higher than those of blank group,the expressions of above indexes of treatment groups wereless than those of model group. From high to low,the expressions of above indexes of treatment groups was 32℃ collapse stains treat-ment group,Diclofenac emulsion group,37℃ collapse stains treatment group and 42℃ collapse stains treatment group.ConclusionsTCM sloughing therapy could reduce the expressions of MMP-3,MMP-13,BMP-2,cylinD1,c-myc mRNA in cartilage of KOA modelrabbits and improve the pathology of articular cartilage of KOA.
