摘要
目的观察胰岛素样生长因子1(IGF-1)对SD大鼠动脉粥样硬化病变处巨噬细胞、TNF-α和血管平滑肌细胞的影响,以探讨其可能的抗动脉粥样硬化机制,为其临床应用提供理论依据和指导。方法 24只8周龄SD大鼠,随机分为模型组(12只)和IGF-1组(12只),分别测量体重、血压、血脂。IGF-1组予高脂饮食+腹腔注射IGF-1 1.0 ml(0.1 mg/ml),模型组予高脂饮食+皮下注射生理盐水1.0 ml,16周后再次测量体质量、血压、血脂,获取两组SD大鼠主动脉行病理分析:HE染色观察横切面动脉粥样硬化程度;免疫组化法半定量观察横切面斑块内TNF-α、巨噬细胞标志物CD68及血管平滑肌细胞标志物α-SMA各自光密度值(OD)。两组间独立样本比较用t检验,以P<0.05为差异有统计学意义。结果与模型组相比,IGF-1组SD大鼠体质量、血压、血脂无显著差异(P>0.05),巨噬细胞标志物CD68及TNF-αOD值显著减少(巨噬细胞OD值:t=6.548,P<0.001;TNF-αOD值:t=6.628,P<0.001),血管平滑肌细胞标志物α-SMA的OD值显著增加(t=12.520,P<0.001)。结论 IGF-1可能通过减少斑块内炎症及炎症因子水平、增加血管平滑肌细胞数量来发挥抗动脉粥样硬化作用。
Objective To observe the effect of the IGF-1 on the lesion of atherosclerosis about SD rats including the number of macrophage,tumor necrosis factor and vascular smooth muscle cells.Furtherly,it could provide the theory and guidance for practise.Methods Twenty-four 8-week-old male SD rats were randomly divided into two groups:the model group(n=12) and IGF-1 group(n=12).To measure the weight,blood pressure and lipid.The IGF-1 group rats were fed with high diet and treated with IGF-1 by intraperitoneal injection,while the model group were fed with high diet and treated with normal saline.After sixteen weeks,measured the weigh,blood pressure and lipid.Laterly,all rats were killed and gained the aortic for pathologic serial section cutting:to observe the lesion of atherosclerosis by HE staining and the number of macrophage and vascular smooth muscle cells,the level of tumor necrosis factor by immunohistochemical,described by optical density.The independent samples between the two groups were compared with t test,and P<0.05 had statistical significance.Results The weight,blood pressure and lipid of rats in the IGF-1 group were not significantly different with that of model group,while compared with the model group,the optical density of macrophage and tumor necrosis factor-α were significantly reduced,while the vascular smooth muscle cells were opposite.Conclusion The effect of IGF-1 on anti-atherosclerosis possibly reduced inflammation level and inflammatory factor,meanwhile,increased the vascular smooth muscle cell.
