芍药苷对H_2O_2诱导的H9C2心肌细胞损伤的保护作用 被引量:3

摘要 目的探讨芍药苷对H_2O_2诱导的H9C2心肌细胞损伤的保护作用。方法培养H9C2心肌细胞,用不同浓度的H_2O_2(50、100、200、400、800、1 600μmol/L)作用于心肌细胞4 h,选出H9C2心肌细胞氧化损伤模型的浓度。以50、100、200μmol/L的芍药苷预处理心肌细胞24 h,以选出的浓度为400μmol/L的H_2O_2建立氧化损伤模型,采用MTT法检测H9C2心肌细胞的活力,化学试剂盒检测心肌细胞乳酸脱氢酶(LDH)漏出量、丙二醛(MDA)、超氧化物歧化酶(SOD)的含量。多组间比较采用单因素方差分析,组间两两比较用LSD检验。结果各H_2O_2浓度组细胞活力显著低于对照组,差异具有统计学意义(P<0.05);各芍药苷浓度组细胞活力高于H_2O_2模型组,且随着芍药苷浓度的升高而升高,差异具有统计学意义(P<0.05);各芍药苷浓度组细胞活力低于对照组,差异具有统计学意义(P<0.05);各芍药苷浓度组细胞LDH漏出量、MDA含量显著低于H_2O_2模型组,且随着芍药苷浓度的升高而降低,差异具有统计学意义(P<0.05);各芍药苷浓度组细胞LDH漏出量、MDA含量高于对照组,差异具有统计学意义(P<0.05);各芍药苷浓度组细胞SOD含量显著高于H_2O_2模型组,且随着芍药苷浓度的升高而升高,差异具有统计学意义(P<0.05);各芍药苷浓度组细胞SOD含量显著低于对照组,差异具有统计学意义(P<0.05)。结论芍药苷对H_2O_2诱导的H9C2心肌细胞的损伤有明显的保护作用。 Objective To investigate the protective effects of paeoniflorin on injury of H9C2 myocardial cells induced by H_2O_2.Methods H9C2 myocardial cells were cultured with different concentrations of H_2O_2(50,100,200,400,800,1 600 μmol/L) for 4 h to select out concentration of oxidation injury model.The myocardial cells were preconditioned with different concentration of paeoniflorin(50,100,200 μmol/L) for 24 h,and then the myocardial cells were cultured with the concentration of H_2O_2 that was 400 μmol/L to establish the oxidative injury model.We used MTT method to detect the activity of H9C2 myocardial cells,and kits to detect lactate dehydrogenase(LDH) leakage quantity,malondialdehyde(MDA),superoxide dismutase(SOD) content of H9C2 myocardial cells.Comparison between the groups used single factor analysis of variance,the comparison between the two groups used LSD test.Results The cell viability of different concentrations of H_2O_2 group was significantly lower than the normal group,the difference was statistically significant(P<0.05); the cell viability of different concentrations of paeoniflorin were significantly higher than that of H_2O_2 model group,and increased with the increase of paeoniflorin concentration,and the difference was statistically significant(P<0.05); the cell viability of different concentrations of paeoniflorin were significantly lowerthan that of control group,and the difference was statistically significant(P<0.05); LDH leakage and MDA content of different concentrations of paeoniflorin were significantly lower than that of H_2O_2 model group,and decreased with the increase of paeoniflorin concentration,and the difference was statistically significant(P<0.05); LDH leakage and MDA content of different concentrations of paeoniflorin were significantly higher than that of control group,and the difference was statistically significant(P<0.05); the SOD content of different concentrations of paeoniflorin was significantly higher than that of model group,and increased with the increase of paeoniflorin concentration,and the difference was statistically significant(P<0.05); the SOD content of different concentrations of paeoniflorin was significantly lower than that of control group,and the difference was statistically significant(P<0.05).Conclusion Paeoniflorin has obvious protective effects on injury of H9C2 myocardial cells induced by H_2O_2.
