摘要
目的观察早期生长反应因子-1(Egr-1)在大鼠腹主动脉瘤发病过程中的表达情况。方法采用弹力蛋白酶灌注法制备大鼠腹主动脉瘤模型,实验组采用猪胰弹力酶灌注,对照组为正常的腹主动脉。14 d后取标本并且测量腹主动脉瘤的直径。应用实时定量聚合酶链反应(q-PCR)、Western blot方法检测动脉瘤中Egr-1 m RNA和蛋白的表达,并对数据进行分析计算。HE染色和透射电镜观察主动脉壁的形态变化来研究观察Egr-1与腹主动脉瘤之间的关系。数据均以t检验进行两组之间的差异分析。结果实验组动物模型均已达到腹主动脉瘤诊断标准(对照组:1.45±0.023,实照组:3.07±0.115,每组10只,P<0.05)。实验组组织形态学显示动脉壁结构紊乱并且有炎性细胞浸润。与对照组相比,实验组Egr-1 m RNA的表达明显高于对照组(对照组:1.00±0.098,实验组:5.74±0.710,P<0.05);Egr-1蛋白的表达明显升高(对照组:0.454±0.054 9,实验组:1.110±0.033 7,P<0.05)。结论 Egr-1在大鼠腹主动脉瘤中的表达明显高于对照组,说明Egr-1可能在介导腹主动脉瘤的形成中发挥重要作用。
Objective To study the early growth response factor-1(Egr-1) variability in abdominal aortic aneurysms(AAA) pathogenesis in rats. Methods Rat infrarenal AAA models were achieved via stretch protease perfusion. Porcine pancreatic elastase was infused in treated group and normal abdominal aorta was in control group. After 14 days, the specimens were taken and the diameter of the abdominal aortic aneurysm was measured. The change of Egr-1 was detected by q-PCR and Western blot. Morphological changes of the aortic wall were observed by HE and transmission electron microscopy. The data were analyzed with means±SD or paired t test for the difference between the two groups. Results The animal model of experimental group had reached the standard of diagnosis of abdominal aortic aneurysm(control group: 1.45±0.023, the experimental group: 3.07±0.115, each group of 10, P<0.05). The experimental group showed the disorder of arterial wall structure. Compared with the control group, the expression of Egr-1 in the experimental group of early growth response factor-1 mRNA was significantly higher than that of control group(control group: 1.00±0.098, the experimental group: 5.74±0.710, P<0.05); and Egr-1 protein expression increased significantly(control group: 0.454±0.054 9, the experimental group: 1.110±0.033 7, P<0.05). Conclusion The changing of Egr-1 expression is parallel to AAA pathogenesis. It suggested that Egr-1 may affect on AAA formation.
