TXNIP/NLRP3炎性小体通路在COPD患者机体炎症反应中的作用研究 被引量:13

摘要 目的:探讨TXNIP/NLRP3炎性小体通路在COPD患者发病机制中的可能作用。方法:选取2017年11月至2018年5月住院的40例COPD患者为急性加重期组,其经过治疗进入稳定期后纳入稳定期组,40例健康体检者为对照组。荧光定量PCR法测定外周血单个核细胞中TXNIP mRNA、NLRP3 mRNA水平,Western blot法检测外周血单个核细胞中TXNIP及NLRP3蛋白表达水平。结果:(1)COPD患者急性加重期组NLRP3、TXNIP的mRNA和蛋白表达水平均显著高于对照组(P<0. 05)和稳定期组(P<0. 05); COPD患者稳定期组的NLRP3 mRNA和蛋白表达水平显著高于对照组(P<0. 05)。(2)相关性分析:急性加重期组NLRP3 mRNA表达量与FEV1%pred、FEV1/FVC负相关;急性加重期组和稳定期组的NLRP3 mRNA、TXNIP mRNA表达量均与CAT评分正相关。结论:TXNIP/NLRP3炎性小体通路参与COPD患者的机体炎症反应。 Objective: To explore the possible role of TXNIP/NLRP3 inflammasome pathway in the pathogenesis of COPD. Methods: 40 cases of COPD patients hospitalized from November 2017 to May 2018 were included in AECOPD group. After the treatment entered the stable period,the patients were included in the stable period group. 40 healthy persons were selected as the control group. The level of TXNIP mRNA and NLRP3 mRNA in peripheral blood mononuclear cells were measured by Real-time PCR,and the expression of TXNIP and NLRP3 protein in peripheral blood mononuclear cells were detected by Western blot. Results:(1) The expression levels of mRNA and protein about NLRP3 and TXNIP in AECOPD patients were significantly higher than those in the control group( P<0. 05) and stable period group( P<0. 05). The expression level of NLRP3 in stable period group was significantly higher than that in control group( P<0. 05).(2)Correlation analysis: The expression of NLRP3 mRNA in AECOPD group was negatively correlated with FEV1% pred and FEV1/FVC. The expression of TXNIP mRNA as well as NLRP3 mRNA in AECOPD group and stable period group were positively correlated with CAT score. Conclusion: The TXNIP/NLRP3 inflammasome pathway is involved in the inflammatory response of COPD patients.
