低次烟叶中绿原酸与芸香苷提取工艺的研究 被引量:3

摘要 旨在获得合适的绿原酸与芸香苷提取工艺,提高低次烟叶的经济附加值。笔者采用单因素试验和响应面设计,高效液相色谱法检测提取量,选择乙醇浓度、料液比、提取时间、提取温度4个因素优化提取工艺。结果表明,绿原酸与芸香苷最佳提取工艺条件为:乙醇浓度67%,料液比1:16 (g:mL),提取温度61℃,提取时间150 min。此条件下,绿原酸与芸香苷理论提取量之和为43.59 mg/g,相对误差0.36%。使用Design-Expert软件对4个因素进行分析,其影响大小顺序依次为:乙醇浓度>提取时间>料液比>提取温度。本研究获得了从低次烟叶中提取绿原酸与芸香苷的工艺条件,为更好利用低次烟叶提供了新思路。 The paper aims to find an appropriate extraction process of chlorogenic acid and rutin from inferior and waste tobacco to improve its economic added value. The authors adopted the single factor experiment andresponse surface design, detected extraction yield by the high performance liquid chromatography(HPLC),optimized the parameters of the extraction process by ethanol concentration, liquid-solid ratio, extraction timeand extraction temperature. The results showed that: the optimized condition for extraction of chlorogenicacid and rutin from inferior and waste tobacco was ethanol concentration of 67%, liquid-solid ratio of 1:16(g:mL), extraction temperature of 61℃ and the extraction time of 150 min. Under this modified condition, thesum of the theoretical extraction yield of chlorogenic acid and rutin was 43.59 mg/g, and the relative errorwas 0.36%. Analysis of the Design-Expert showed that: ethanol concentration>extraction time>liquid-solidratio>extraction temperature. The technological condition of extracting chlorogenic acid and rutin frominferior and waste tobacco obtained by the study could provide a new way for better utilization of waste tobacco.
