人胃癌移植瘤裸小鼠Ki-ras基因突变的检测

Detection on mutation of Ki-ras gene of human gastric cancer xenografts in nudemice

目的 检测人胃癌裸小鼠移植瘤Ki ras基因的突变情况。方法 将人胃癌细胞SGC 790 1于裸小鼠脾包膜下移植 ,12周后处死 ,采用PCR DNA测序法检测Ki ras基因第 12、13密码子的突变情况。结果 共检测 2 9例裸鼠移植瘤 ,DNA经扩增后 2 0例为阳性 ,其中 12例进行DNA测序 ,未发现第 12、13密码子的突变。结论 人胃癌移植瘤裸小鼠Ki ras基因第 12。

Objective To detect the mutation of Ki ras gene exon 12, 13 of human gastric cancer xenografts in nude mice. Methods A human gastric adenocarcinoma cell line SGC 7901 was grafted under splenic capsule of 32 mice, Ki ras gene exon 12, 13 were detected by PCR DNA method 12 weeks after the procedure. Results No Ki ras gene exon 12, 13 mutations were detected. Conclusions The presence of mutation of Ki ras gene exon 12, 13 on human gastric cancer xenografts of nude mice can not be confirmed.
