PDTC对过氧化氢诱导鼠晶状体上皮细胞NF-κB活化表达的影响 被引量:9

摘要 目的 观察过氧化氢 (H2 O2 )诱导鼠白内障形成过程中晶状体上皮细胞核因子 KB(NF κB)的活化表达及吡咯烷二硫氨基甲酸 (PDTC)对核因子κB(NF κB)活化表达的抑制作用 ,探讨NF κB及PDTC在白内障发生、预防和治疗中的作用。方法 大鼠晶状体器官离体培养 ,免疫组织化学方法检测晶状体上皮细胞NF κB的活化表达。结果 随过氧化氢损伤时间的延长 ,H2 O2 组晶状体上皮细胞NF κB活化表达逐渐增强 ,且与晶状体混浊程度呈正相关。PDTC可以明显抑制晶状体上皮细胞NF κB阳性活化表达和减轻晶状体混浊程度 ,且存在一定程度的剂量依赖性。结论 过氧化氢可以诱导鼠晶状体上皮细胞NF κB的活化表达 ,PDTC可以有效抑制NF κB的活化表达 。 Objective To study the active expression of nuclear5 factor κB(NF κB) and the inhibitory role of pyrroledithiocarbomate(PDTC) on NF κB during H 2O 2 induced cataract formation.And to investigate action of NF κB and PDTC in cataract pathogenesis and prevention and therapy.Methods After the vitro culture of rat's lens organ,we examined the active expression of NF κB in rat lens epithelial cell by using immunohistochemistry technique.Results The longer incubated with H 2O 2 treatment,the more active expression of NF κB in lens epithelial cell were found,the latter was positively correlated with opaque degree of lens and to a certain extent PDTC had a dose dependent action in inhibiting the active expression of NF κB in rat's lens epithelial cells and relieve opacity of lens.Conclusion The active expression of NF κB in rat's lens epithelial cell was induced by H 2O 2,and PDTC could effectively inhibit the active expression of NF κB in rat's lens epithelial cells,and might confront or postpone the occurrence of cataract.
