摘要
高质量的DNA是林木分子生物学研究的关键因素之一。在高浓度离液剂异硫氰酸胍(GuSCN)存在的条件下,核酸具有与硅珠(SiO2)颗粒结合、并能在较高温度下被低盐浓度溶液洗脱的特性。基于这一原理建立了一种新的树木叶片总DNA提取方法。此方法操作简单、快速,经过浓度、纯度测定,以及PCR扩增和限制性内切酶消化证明,从不同树木叶片中均得到了高质量的DNA样品。
High quality DNA samples are one of key factors in study of forest molecular biology.Based on the presence of high concentrations of the chaotropic agent guanidinium thiocyanate(GuSCN),the nucleic acid could bind to silica particles(SiO_2) and subsequently was eluted in a aqueous low-salt buffer.A new method of total DNA extraction from leaves of tree was established.The method is rapid and simple.High quality DNA could be isolated from leaves of (5 tree) species proved by the concentrations,purities,PCR amplification and restriction digests.This method could be used widely in study of forest molecular biology.
