摘要
目的 探讨p73基因转染对肺腺癌细胞体外生长的抑制作用及 p73基因治疗肺腺癌的可能性。方法 利用脂质体将p73β基因转导入两株分别对 p5 3基因治疗敏感和耐受的肺腺癌细胞系H1 2 99(p5 3 null)和A5 4 9(wtp5 3)中 ,对 p73蛋白过表达的细胞进行细胞生长曲线、克隆形成率分析 ,并用流式细胞术分析p73β基因对肺腺癌细胞周期的影响和细胞增殖的抑制作用。结果 导入p73β基因能使H1 2 99和A5 4 9细胞发生G1 期阻滞 ,生长速度明显减慢 ,克隆形成率下降。结论 外源性p73β基因转染可以抑制肺腺癌细胞体外生长 ,且这种抑制作用与p5 3基因无关。
Objective To investigate the suppression effect of exogenous p73 gene on human lung adenocarcionoma cell lines and the possibility of p73 gene transfection for gene therapy of lung cancer.Methods p73β expression vector was transfected into p53 sensitive lung adenocarcionoma cell line,H1299,and p53 resistant lung adenocarcionoma cell line,A549.The suppression effect of p73 gene on cell growth was detected by cell growth curve and cloning efficiency assay.The cell cycle pattern was assessed by flow cytometry.Results The growth and colony forming efficiency of H1299 p73β and A549 p73β cells were significantly inhibited compared with the controls.Flow cytometry showed H1299 p73β and A549 p73β were arrested in G1 phase of cell cycle.Conclusions Exogenous p73β gene can inhibit proliferation and induce apoptosis of p53 sensitive and p53 resistant lung adenocarcionoma cell lines.Therefore,the results suggest that the p73β gene may be useful in cancer treatment.
