Effects of Osmoconditioning on the Raising of Vigor and the Metabolism of Nucleic Acid in Peanut Seeds

摘要 应用20%PEG 6000(聚乙二醇)处理花生种子,可以促进萌发,增加幼苗生长量.种子经6小时PEG 处理,总^(32)P 吸收量和掺入RNA 的^(32)P 水平均有所增加,可见播前PEG 处理可促进与萌发有关的过程,“RNA 掺入^(32)P 量/总^(32)P 吸收量”比值以PEG 处理2天的为最高,3—5天处理的反而有所下降,从种子萌发效果看,PEG 处理2天是较适时间. Peanut seeds,conditioned with 20% PEG at 17℃ for two days followed byair drying at 30℃ for two hours,germinated rapidly and had greater length of radicle-hypocotyl.The osmoconditioning advanced in parellel fashion the rateof phosphate uptake and RNA synthesis in peanut seeds as measured by the timecourse of NaH_2^(32)PO_4incorporation into the phosphate compounds or RNA.Theincrease of ^(32)P-uptake and RNA synthesis began at about 6h or more afterthe osmotic treatment.There was the highest ratio of ^(32)P-RNA/^(32)P-total duringosmoconditioning for two days,while for 3-5 days the ratios were lowereddown.It was interested to note that the effect of osmoconditioning for two dayswas the best on accelerating the germination rate and increasing the seed vigor.The incorporation of ^(32)p into RNA and other phosphate compounds was higherin the seeds conditioned with PEG for two days than that in the control seeds.During the osmoconditioning period or period of germination after PEG treatment,the changes of DNA were inconspicuous,the amounts of DNA synthesized inthose seeds were also very little.
