辽宁地区野生和栽培羊肚菌ITS序列及ISSR多态性分析

Analysis on ITS Sequence and ISSR Polymorphism of Wild and Cultivated Morchella in Liaoning Province

针对采集到的15株栽培羊肚菌样品和14株野生羊肚菌样品进行ITS序列分析和ISSR多态性分析,通过构建系统发育树和遗传聚类分析图谱,发现辽宁地区主栽品种为黑色系的六妹羊肚菌(Morchella sextelata,Mel-6)和梯棱羊肚菌(Morchella importuna,Mel-10),同时还有少量的七妹羊肚菌(Morchella septimelata,Mel-7);野生羊肚菌样品属于黄色系的Morchella sp.(Mes-6)和Morchella sp.(Mes-9)。结果表明辽宁地区羊肚菌栽培品种与野生羊肚菌品种具有明显的遗传差异性,为实现本地野生羊肚菌资源的合理利用和人工栽培研究提供参考。

15 strains of cultivated Morchella sp. and 14 samples of wild Morchella sp. were collected, which were analyzed by ITS sequence analysis, ISSR polymorphism and construction of phylogenetic trees and genetic clustering analysis maps. The results showed the main varieties in Liaoning province were Morchella sextelata(Mel-6) and Morchella importuna(Mel-10), and there were a small amount of Morchella septimelata(Mel-7). The wild Morchella sp. samples belong to yellow strain were Morchella sp.(Mes-6) and Morchella sp.(Mes-9). Genetic differences was obvious between cultivated Morchella and wild varieties in Liaoning province, which provide reference for the rational utilization of wild morel resources and artificial cultivation research.
