摘要
观察射干等20种中药的体外抗真菌活性。通过乙醇回流、水煎煮和超声波等方法提取射干等20种中药中的抗菌活性部位,采用液体微量稀释法(broth microdilution susceptibility method,M38-A)测定射干等20种中药对石膏样小孢子菌、犬小孢子菌、须癣毛癣菌和白色念珠球菌4株真菌的体外抗菌活性,并研究抗菌活性较强几味中药的体外联合抑菌活性。射干、白鲜皮、五倍子和苦参4种中药提取物对4株真菌的最低抑菌浓度(minimal inhibitory concentration,MIC)范围为1.563~12.5g/L;姜黄、杜仲、赤芍、栀子、连翘和柴胡6种中药提取物的MIC值范围为6.25~50g/L;藿香、黄芩、板蓝根、黄柏和大黄5种中药提取物的MIC值范围在50~100g/L;金银花、虎杖、何首乌、知母和佛手5种中药提取物的MIC值>100g/L。联合抑菌试验结果表明,射干、姜黄和栀子的联合抑菌指数(FICI)≤2,五倍子和射干、姜黄、栀子的联合抑菌指数(FIC)>2。结果显示射干、五倍子、姜黄、栀子对4株真菌均具有较好的体外抗真菌活性;射干、姜黄、栀子表现出无关或相加、协同的作用;五倍子和射干、姜黄、栀子表现出了拮抗作用。
The in vitro anti-fungal activities of twenty kinds of traditional Chinese medicine were evaluated in this study.The anti-fungal active sites of twenty kinds of traditional Chinese medicine like Belamcanda chinensis were extracted by the methods of ethanol reflux extraction,water decoction,ultrasonic wave and so on.The anti-fungal activities against Microsporum gypseum,Microsporum canis,Trichophyton mentagrophytes and Microsporic mycosis were determined by the broth microdilution susceptibility method(M38-A).And then the joint bacteriostatic activities of the medicine were evaluated in vitro.The results showed that the MIC of the extracts obtained from Belamcanda chinensis,Dictamnus dasycarpus Turcz,Melaphis chinensis and Sophora flavescens Ait against four strains of fungi were a range from 1.563g/L to 12.5g/L;MIC of the extracts obtained from Curcuma longa L,Fucommia ulmoides Oliv,Paeonia lactiflora Pall,Gardenia jasminoides Ellis,Forsythia suspensa(Thunb.)Vahl and Bupleurum chinense DC were a range from 6.25g/L to 50g/L;the MIC of the extracts obtained from Scutellaria baicalensis Georgi,Phellodendron chinense Schneid,Isatis indigotica Fort,Rheum palmatum L and Agastache rugosus were a range from 50g/L to 100g/L;the MIC > 100g/L included Lonicera japonica Thunb,Polygonum cuspidatum Sieb.et Zucc,Anemarrhena asphodeloides Bge,Polygonum multiflorum Thunb and Citrus medica L,var sarcodactylis Swingle.The results of joint bacteriostatic test showed that the fractional inhibitory concentration index(FICI)of Belamcanda chinensis,Curcuma longa L and Gardenia jasminoides Ellis was≤2and FICI of Melaphis chinensis and Belamcanda chinensis,Curcuma longa L,Gardenia jasminoides Ellis were>2.Belamcanda chinensis,Melaphis chinensis,Curcuma longa L and Gardenia jasminoides Ellis had good antimicrobial activities against four kinds of fungi.Melaphis chinensis and Belamcanda chinensis,Curcuma longa L,Gardenia jasminoides Ellis showed antagonistic action and Belamcanda chinensis and Curcuma longa L,Gardenia jasminoides Ellis showed independent,additive or synergy action.
