摘要
快慢羽性状为伴性遗传性状,可通过建立雌雄自鉴别品系应用于家禽的早期性别鉴定。因慢羽基因对快羽基因呈显性,故区分慢羽公鸡的羽速基因型是建立慢羽纯系的重要工作。使用传统测交鉴定方法鉴别耗时费力,而已有的分子鉴定方法有的成本太高,有的准确率太低。本研究以已有表型记录的快羽或慢羽太行鸡公母鸡为试验材料,设计了半定量PCR鉴定公鸡慢羽羽速基因型引物,通过摸索最佳反应体系和条件并通过对测交验证,最终建立了能较准确鉴定太行鸡慢羽公鸡羽速基因型方法,为太行鸡慢羽纯系的建立奠定技术基础。
Slow and fast feathering traits are sex-linkage traits and could be used to identify the sex of poultry at early age.The slow feathering gene is dominant to the fast feathering one.So to authenticate the genotype of late featering male chicken is an important work for establishing late feathering chicken lines.It is time-consuming and laborious to identify the feathering-speed genotype of the slow-feather cock by using the method of traditional test cross.Therefore,Using the chickens known the feathering phenotypic(slow-or fast-feathering)as experiment material and refered to the related literature,semi-quantitative PCR primers were designed for identification feathering speed genotype.Through groping for the best reaction system and reaction conditions,a semi-quantitative PCR method was setted up and the accuracy of this method was tested by test cross.This research laid a solid foundation for the establishment of the late-feathing line of taihang chicken.
