摘要
目的探讨miR-155对结肠癌细胞增殖和线粒体凋亡信号通路的影响,为明确miR-155在结肠癌发生发展中的作用提供理论依据。方法收集临床结肠癌以及癌旁组织,体外培养人结肠癌细胞,RT-qPCR检测组织中miR-155表达,结肠癌细胞给予miR-155抑制剂,CCK8检测细胞增殖率的改变,流式细胞术检测凋亡率的改变,Western blotting检测线粒体凋亡相关蛋白的表达。结果与癌旁组织相比,结肠癌组织中miR-155表达水平明显升高,miR-155抑制剂可以明显的降低结肠癌细胞增殖率,诱导凋亡的发生;同时可以增加线粒体凋亡相关蛋白Bax和cytochrome C表达。结论 miR-155在结肠癌组织中呈现高表达,抑制miR-155可以明显的降低结肠癌细胞的增殖率,诱导线粒体凋亡信号通路的活化。
Objective To explore the effects of miR-155 on the proliferation and mitochondrial apoptosis signaling pathways of colon cancer cells,providing a theoretical basis for clarifying the role of miR-155 in the development and progression of colon cancer.Methods Clinical colon cancer and adjacent tissues were collected,human colon cancer cells were cultured outside,miR-155 expression was detected by RT-PCR,colon cancer cells were given miR-155 inhibitors,cell proliferation rate was detected by CCK8,apoptosis rate was detected by flow cytometry,and expression of mitochondrial apoptosis-related proteins was detected by Western blotting.Results compared with the paracancerous tissue,the expression level of miR-155 in colon cancer tissue was significantly increased,and the miR-155 inhibitor could significantly reduce the proliferation rate of colon cancer cells and induce apoptosis.Meanwhile,the expression of cytochrome C and Bax related to mitochondrial apoptosis was increased.Conclusion miR-155 is highly expressed in colon cancer tissues.Inhibition of miR-155 can significantly reduce the proliferation rate of colon cancer cells and induce the activation of mitochondrial apoptosis signaling pathway.
