大黄总蒽醌的提取纯化及含量测定 被引量:4

摘要 目的:提取纯化大黄药材中的总蒽醌类成分,使其纯度达到有效部位的要求。方法:参考《中国药典》(2010年版)方法,先用75%乙醇提取,将所得干膏用8%盐酸水解-氯仿萃取后,用D301大孔树脂纯化处理,用紫外-可见分光光度法测定含量。结果:大黄总蒽醌以大黄素计,在0.000 8-0.020 0 mg/m L范围内线性关系良好,回归方程为:A=42.039 C-0.010 4(r=0.999 2)。经提取纯化,大黄总蒽醌纯度达80%以上。结论:用此方法得到的大黄蒽醌纯度较高,达到有效部位的要求。 Objective: To extract and purify total anthraquinones in Rhubarb, and make it meet the requirements of effective fraction. Methods: Radix et Rhizoma Rhei was extracted with 75% of ethanol, and the dry extract was hydrolyzed by 8% of hydrochloric acid, extracted by chloroform, purified by D301 macroporous resin, and determined by ultraviolet-visible spectrophotometry. Results: The calibration curve of total anthraquinones showed good linearity in the range of 0.000 8-0.0200mg/m L, and the regression equation was A=42.039C-0.0104(r=0.999 2). The average recovery of the method was 95.83% with corresponding RSD of 1.79%. After extraction and purification, the purity of total anthraquinones in Rhubarb was more than80%. Conclusion: High purity of total anthraquinones in Rhubarb obtained with this way meet the need of the effective parts.
