不同熏硫剂量对当归组织结构、pH值及阿魏酸含量的影响 被引量:1

Effects of different doses of sulfur fumigation on the tissue structures, pH and ferulic acid of Angelica sinensis

摘要 目的:探讨不同熏硫剂量对当归组织结构、pH值及阿魏酸含量的影响,为提高中药材传统加工品质提供科学依据。方法:分别收集新鲜当归和市售熏硫当归样品,对新鲜当归药材分别以高剂量(5.0%)、中剂量(1.0%)、低剂量(0.2%)3个不同剂量进行熏硫加工;采用显微技术对其油室(分泌腔)形态、数目进行观察;通过pH计测定其浸出物的pH值;采用HPLC测定其阿魏酸的含量。结果:熏硫后当归外观色泽变浅,组织结构发生变化,单位面积(mm^2)上油室大小、数目及分布均显示出一定差异。随着熏硫剂量的增加,单位面积上油室直径增大、油室数量减少、油室破裂较多。熏硫后当归浸出物pH值降低,5%熏硫剂量时当归浸出物pH值降低最为明显(21.98%)。当归中阿魏酸含量随熏硫剂量增加先升高后降低,1%熏硫剂量时当归中阿魏酸含量增加最为明显(11.88%)。结论:熏硫对当归药材有一定的影响,为保证当归药材质量和临床用药安全,产地加工应严格控制熏硫剂量。 Objective:To explore the effects of different doses of sulfur fumigation on the tissue structures,pH and ferulic acid of Angelica sinensis,and provide scientific basis for ensuring the quality of authentic Chinese medicinal materials in Gansu.Methods:Fresh and commercially available samples of sulfur fumigation and Angelica sinensis were collected separately,and the fresh Angelica sinensis medicinal materials were subjected to fumigant processing in three doses of high(5%),medium(1%)and low(0.2%),respectively.The tissue structures were observed and the content of ferulic acid in different sulphur dosages of Angelica sinensis were determined by microscope and high performance liquid chromatography.Results:After sulfur fumigation,the color gradually became lighter of Angelica sinensis.Tissue structures have also been changed.After sulfur fumigation,the diameter of the oil chamber became larger,the number of oil chambers per unit mm2 were reduced,and the oil chamber were ruptured more.After sulfur fumigation of Angelica sinensis,the pH of extract was lowered,especially with 5%sulfur smoked dose most significant(21.98%).The content of ferulic acid in Angelica sinensis increased firstly with increase of the dose of sulfur fumigation,and the dose of 1%sulphur was the most obvious(11.88%).Conclusion:Sulfur fumigation had certain influence on Angelica sinensis.In order to ensure the quality of Angelica sinensis and the safety of clinical medication,the processing of production should strictly control the dose of sulfur fumigation.
