镁合金微弧氧化掺杂HA硅烷/植酸复合膜细胞相容性研究 被引量:1

摘要 目的为了提高镁合金的细胞相容性和耐腐蚀性,通过对镁合金表面进行微弧氧化-硅烷/植酸处理后,掺杂不同浓度的HA,研究对成骨细胞生物相容性的影响。方法选用MC3T3-E1小鼠成骨细胞,以镁合金微弧氧化-硅烷/植酸组为对照A组,分别添加2 g/L、4 g/L、6 g/L HA的试件为B、C、D实验组。通过SEM观察试件表面形貌、检测表面元素,CCK-8实验检测不同时间下细胞黏附情况以及增殖情况,激光共聚焦检测早期细胞骨架形态。结果 CCK-8检测结果显示,四组试件表面细胞的增殖及黏附顺序为C组优于D组优于B组优于A组,其中C组具有最优的成骨细胞相容性。据统计学分析四组实验数据均具有统计学意义。结论镁合金微弧氧化-硅烷/植酸载HA处理后涂层可增强试件的细胞相容性。当HA的浓度达4 g/L时,涂层具有最优的细胞相容性。 Objective In order to improve the corrosion resistance and cellular biocompatibility of magnesium alloy,the effects of HA on the biocompatibility of osteoblasts were studied by micro-arc oxidation-silane/phytic acid treatment on the surface of magnesium alloys.Methods MC3 T3-E1 mouse osteoblasts were selected.The magnesium alloy micro-arc oxidation-silane/phytic acid group was used as the control group A.The groups B,C,D are with 2 g/L,4 g/L,6 g/L HA added respectively.The surface morphology and surface elements of the specimens were observed by scanning electron microscopy.CCK-8 was used to detect cell adhesion and proliferation at different times.Laser confocal microscopy was used to detect the morphology of early cytoskeleton.Results Results of CCK-8 tests showed that Group C has the best cellular biocompatibility.It has optimal cell proliferation,adhesion,and cytoskeletal extension.Group A is the worst.While group B and D is between these two groups.According to statistical analysis,the experimental results are statistically significant.Conclusions Microarc oxidation-silane/phytic acid-HA coatings on magnesium alloys can enhance the cell compatibility of the specimens.The coating has optimal cellular biocompatibility when the concentration of HA is 4 g/L.
