摘要
目的:在反相条件下建立佐匹克隆对映体的简单、快速的拆分方法。方法:使用自制的未衍生β-环糊精手性固定相,以甲醇-水(或醋酸钠-醋酸缓冲液)为流动相,研究了流动相的组成、pH、柱温、流速对分离的影响,确定最佳色谱条件为流动相:甲醇-水(40:60),柱温为25℃,流速为0.2 mL·min-1,紫外检测波长为305 nm。结果:在所建立的色谱条件下,佐匹克隆对映体接近基线分离,探讨了拆分的机理,并与CHIRAL OD-H进口柱的拆分效果进行了比较。结论:所建立的方法直接、简便、快速。
Objective: To develop a simple, rapid separation method for the enantiomers of zopiclone, a novel drug for the short - term treatment of insomnia, transient, situational or chronic insomnia. Method: Using β - CD chiral column treated by the authors, methanol -water (or NaAc - HAc buffer solution) as mobile phase, 305 nm as the detective wavelength, the influences of the composition of mobile phase, pH, column temperature and flow rate on the enantiomeric separation of zopiclone were investigated and the optimal chromatographic conditions for the separation were established. Results: Under the chromatographic conditions adopted, nearly baseline separation could be obtained. Based on the experimental data, the mechanism of separation was discussed. Conclusion: The proposed method can be applied to the assay of zopiclone in reverse phase conditions easily and rapidly.
