摘要
研究了免疫电镜技术在新疆哈密瓜病毒病诊断中的应用。预先用黄瓜花叶病毒(CMV)、西瓜花叶病毒—2(WMV—2)和南瓜花叶病毒(SqMV)的抗血清处理的电镜载网可以分别捕获到大量的CMV、WMV—2和SqMV的病毒粒子。不同抗血清处理的电镜载网对病毒粒子的捕获具有严格的专一性。抗血清处理过的电镜载网可以贮存于4℃冰箱中达1个月之久仍然具有良好的捕获病毒粒子的能力。一个样品的全部检测程序可以在约30分钟内完成。这些结果肯定了免疫电镜技术在检测CMV、WMV—2和SqMV时是有效的,并为进一步研究新疆哈密瓜病毒病的流行规律提供了迅速、准确的手段。
The study of applicating immune electron microscopy in the diagno-sis of Xinjiang Hami muskmelon virus disease was carried out.The pre-coated electron microscopic grids with antisera against CMV,WMV-2 andSqMV could trap a lot of particles of CMV,WMV-2 and SqMV respec-tively.Grids coated with different antiserum could only trap relevantVirus particles.The coated grids with antiserum could still maintain bet-ter ability to trap virus particles even by storing in 4℃ condition for4 weeks.The whole procedure to diagnose a sample could be finishedin about 30 minutes.The results indicate that the immune electron microscopy is very sen-sitive for detecting and diagnosing CMV,WMV-2and SqMV.This studyprovides a rapid and accurate method for studying epidemic pattern ofXinjiang Hami Muskmelon virus disease.
