摘要
目的探讨棉酚联合榄香烯对Lewis肺癌细胞的抑制效应,为肺癌的治疗提供新的治疗依据。方法以不同浓度的棉酚、榄香烯单独及联合作用于Lewis肺癌细胞株(LLC),在24h末,ATP溶液(Caymen Chemicals,St Louis,MO)添加在细胞中,应用ATP水平测定板测定LLC细胞的ATP表达水平。分别测定单纯LLC细胞、单纯应用棉酚、单纯应用榄香烯以及选取合适的浓度进行联合用药后对LLC细胞ATP表达水平的影响,对ATP数据分别进行计算,分析判断药物对肿瘤细胞生长的影响。结果榄香烯浓度为5.0μg/ml对Lewis肺癌细胞的抑制率(57.3%)效果明显高于2.5μg/ml(20.4%),差异有统计学意义(P <0.05)。棉酚浓度为3.0μg/ml和4.0μg/ml时,其对Lewis肺癌细胞的抑制效果明显高于其他浓度,有显著统计学差异;特别是当棉酚浓度为4.0μg/ml对Lewis肺癌细胞的抑制效果较佳,为73.2%。显著高于1.0μg/ml(21.3%),2.0μg/ml(63.2%),3.0μg/ml(70.3%)。而棉酚浓度为3.0μg/ml时对Lewis肺癌细胞的抑制率(70.3%),显著高于1.0μg/ml(21.3%),2.0μg/ml(63.2%)。棉酚浓度为2.0μg/ml时对Lewis肺癌细胞的抑制率(63.2%),显著高于1.0μg/ml(21.3%),差异均有统计学差异(均P <0.05)。榄香烯浓度为2.5μg/ml时,联合棉酚浓度3.0μg/ml抑制率为(89.6%)和4.0μg/ml抑制率为(92.0%)时,其效果明显高于棉酚浓度1.0μg/ml抑制率为(21.6%)和2.0μg/ml抑制率为(80.1%),差异均有统计学差异(均P <0.05)。特别是榄香烯联合棉酚浓度为4.0μg/ml抑制率为(92.0%),对Lewis肺癌细胞的抑制效果最佳,明显高于其他浓度组。榄香烯浓度为5.0μg/ml时,联合棉酚浓度3.0μg/ml抑制率为(81.1%)和4.0μg/ml抑制率为(90.0%)时,其效果明显高于棉酚浓度1.0μg/ml抑制率为(69.2%)和2.0μg/ml抑制率为(76.2%),差异均有统计学差异(均P <0.05)。特别是榄香烯联合棉酚浓度为4.0μg/ml抑制率为(90.0%),对Lewis肺癌细胞的抑制效果最佳,明显高于其他浓度组。结论棉酚、榄香烯单独或二者联合作用均能抑制LLC的增殖,且联合应用的作用显著高于单独用药,同时能够减少常规榄香烯的用量。
Objective To investigate the Inhibitory Effect of Gossypol and Elemene injection on Lewis lung cancer cell.Methods Lewis lung cancer lines were treated with different concentrations of Gossypol and Elemene injection and combination of them separately.At the end of 24 hours,ATP solution(Caymen Chemicals,St Louis,MO)were added to the wells,the level of ATP were measured with plate reader(Biotek Inc,Bellirica,MA).Data of cells cultured in the presence or absence of Gossypol and B-Elemene were calculated and presented.Results(1)Cell proliferation was dramatically inhibited by Gossypol or Elemene of injection different concentrations,but the inhibitive rate was not higher.(2) The inhibition rate of drμg combination On LCC was higher than that of monotherapy.The inhibitiory rate of 4μg/ml Gossypol and 2.5'https://cn.binjection on Lewis lung cancer cell lines was 92.Conclusion Gossypol,Elemene injection showed the inhibitory effect on Lewis lung cancer cell lines distinctly,and the two drμgs have better cooperative effect when combinated,for the more,the Elemene injectionene and the two drugs ha.
