Evaluation of the Content of Flax Lignan and Genetic Diversity

摘要 用高效液相色谱法对50份亚麻种质木酚素含量检测,并对其进行基于SRAP标记的遗传多样分析,进一步了解亚麻种质的木酚素含量变异和遗传多样性,为高木酚素含量品种选育和后续亚麻木酚素品质改良育种提供科学依据。结果表明,供试材料的木酚素含量变幅为2.819~13.001μg/g,平均值为7.581μg/g,最高木酚素含量种质为加拿大的J-309。11对SRAP引物扩增得到135个条带,多态性条带为73,多态性比率为54.07%,引物多态性信息量(PIC)平均为0.74。有效等位基因数(N_e)、香农信息指数(I)和Nei′s遗传相似系数(H)分别为1.6010、0.7341和0.4925。木酚素含量和基因型数据的聚类分析结果表明,两种不同聚类的同一个类群中大部分种质相同。亚麻种质之间的木酚素含量差异和遗传多样性受地域影响较大。 The lignan content of 50 flax germplasms was determined by high performance liquid chromatography andsubjected to genetic diversity analysis based on SRAP markers. The purpose of our study was to further understand thelignan content variation and genetic diversity of flax germplasm and provide a scientific basis for the breeding of highlignan content and the subsequent improvement of flax lignan quality. The results showed that the lignan content of thetested materials ranged from 2.819 to 13.001μg/g, with an average of 7.581μg/g, and the highest lignan content was J-309 from Canada. 11 pairs of SRAP primers were amplified. There were 135 bands, the polymorphic band was 73, thepolymorphism ratio was 54.07%, and the primer polymorphism information(PIC) averaged 0.74. The effective allelenumber(N_e), the shannon′s information index(I), and the Nei’s genetic similarity coefficient(H) were 1.6010, 0.7341,and 0.4925, respectively. The results of cluster analysis of lignan content and genotype data show that the two clustersare divided into three categories. Most of the same species in the two different clusters are identical. It is concluded thatthe difference in lignan content and genetic diversity between flax germplasms are significantly affected by the region.
