β-淀粉样蛋白对神经干细胞致凋亡作用研究

Study on effects of β-amyloid protein on cultured neural stem cell in vitro

目的观察β-淀粉样蛋白(Aβ)能否诱导体外培养的大鼠神经干细胞凋亡。方法取孕13d的胎鼠大脑,体外进行培养、传代、鉴定后,加入Aβ,利用细胞计数观察细胞增生,利用琼脂糖凝胶电泳及流式细胞仪观察凋亡情况。结果当Aβ浓度>25μmol/L时,能明显抑制神经干细胞的增生,同时引起神经干细胞的明显凋亡。结论Aβ抑制神经干细胞的增生并可致神经干细胞凋亡,可能与阿尔茨海默病的发病有关。

Objective To study the effects ofβ-amyloid protein(Aβ)on neural stem cell(NSC)cultured in vitro.Methods Neural stem cell(NSC)was isolated from E13SD rat and cultured in serum-free medium(DMEM/F12).After detected by nestin immsunofluorescent staining,Aβwas added to the NSC medium,then proliferation of NSC was observed by cell counting and apoptosis by agarose gel electrophoresis and flow cytometry.Results Aβmarkedly inhibi-ted the proliferation of NSC and induced apoptosis when its concentration was higher than25μmol/L.Conclusion Aβcan significantly inhibit the proliferation of NSC and induce its apoptosis,which may be one of the causes of Alzheimer disease.
