乌参醒脑方对6-羟基多巴胺诱导帕金森病大鼠的改善作用 被引量:3

The therapeutic effect of Wushen-Xingnao Fang on 6-hydroxydopamine-induced Parkinson's disease

摘要 目的:观察乌参醒脑方(WSXN)对6-羟基多巴胺(6-OHDA)诱导帕金森病(PD)模型的治疗作用。方法:雄性SD大鼠纹状体注射6-OHDA建立PD模型,分假手术组、模型组、多巴丝肼组、WSXN(22. 5,45和90 mg·kg^(-1))剂量组。给药24 d,计算APO激发转圈频率和损侧纹状体/脑指数,测定纹状体DA含量和血清SOD、MDA和GSH-Px水平;尼氏染色观察纹状体神经元功能和免疫组化检测黑质多巴胺能神经元数目。结果:与模型组相比,WSXN各剂量组转圈频率减少;纹状体/脑指数升高、DA含量增加,神经元功能改善; SOD和GSH-Px活力上升,MDA水平下降; DA能神经元丢失减轻。结论:WSXN对6-OHDA诱导PD大鼠有一定的治疗作用,其作用可能与抗氧化应激水平、减轻纹状体损伤,减少多巴胺能神经元丢失和提高脑内多巴胺含量有关。 Objective: To investigate the effect of Wushen-Xingnao Fang( WSXN) on 6-OHDA-induced Parkinson’s disease. Methods: Male SPF SD rats were selected to establish PD model by injecting 6-OHDA in striatum and divided into model group,madopar group,WSXN( 22. 5,45,90 mg·kg-1) groups and sham group.After drug administration for 24 days,the circling frequency and striatum/brain index were calculated. The total SOD,MDA and GSH-Px in serum and the content of DA in striatum were determined. Nissl staining and immunohistochemistry were used to evaluate the neuronal function and the number of DA neurons in midbrain. Results:The circling frequency in WSXN groups was significantly reduced,compared with the model group. The striatum/brain index,DA content and neuronal function were improved or increased obviously. There was an elevation of the total SOD and GSH-Px,while decline in MDA level and a significant improvement of DA neurons in the WSXN groups. Conclusion: WSXN has therapeutic effect to improve the behavioral symptoms in PD rats induced by 6-OHDA,and its mechanisms may be associated with reduction of oxidative stress and damage of striatums,and increasing of DA content in midbrain.
