基于PB设计和BBD响应面法优化制天南星多糖超声辅助提取工艺及抗肿瘤活性研究 被引量:12

Optimization of ultrasound-assisted extraction technology based on PB design and BBD response surface method and anti-tumor activities of polysaccharides from Arisaematis rhizoma preparatum

摘要 目的:利用Plackett-Burman和Box-Behnken效应面法优化制天南星的多糖超声提取工艺,并研究其抗肿瘤活性。方法:采用Plackett-Burman设计实验,以制天南星多糖的提取率为指标,筛选影响多糖提取率的5个影响因素,在此基础上,通过Box-Behnken响应面法对筛选出的显著影响因素进一步优化制天南星多糖提取工艺,同时观察制天南星多糖的体外抗肿瘤作用。结果:优化后的最佳工艺为粉碎粒径80目,液料比1∶30(g·m L^(-1)),超声时间50 min,超声温度58℃,超声功率210 W,在此条件下制天南星多糖提取率为9. 12%,对人肺癌A549的抑制率IC50为8. 625 mg·m L^(-1)。结论:采用PB设计和BBD响应面法优化制天南星多糖提取工艺稳定可行。 Objective:The Plackett-Burman and Box-Behnken effect surface method were used to optimize the ultrasound-assisted extraction technology of arisaematis rhizoma preparatum polysaccharides(ARPP)and its antitumor activity was studied.Methods:Basis on single factor experiment,five factors affecting the extraction of ARPP were screened by Plackett-Burman(PB)design.On this basis,the Box-Behnken response surface method was used to optimize the polysaccharide extraction process of ARPP.And the survival rate of A549 cell were used to evaluate antitumor activity of ARPP.Results:Results indicated that the optimal extraction conditions were as follows:particle size was 80 meshes,the ratio of water to raw was 30 m L·g-1,extraction time was 50 min,ultrasonic temperature was 58℃,and ultrasonic power was 210 W.Under these conditions,the extraction rate of ARPP was 9.12%,and the inhibitory rate IC50 on human lung cancer A549 was 8.625 mg·m L-1.Conclusion:The extraction process of ARPP optimized by PB design and Box-Behnken effect surface method is effective and feasible with a good prediction.
