抑酸药物与食物对小分子靶向药物口服吸收的影响

Effects of antiacid drugs and food on the oral absorption of small molecular targeted drugs

摘要 抑酸药物(antiacid drugs,AD)和食物可影响一部分小分子靶向药物(small molecular targeted drugs,SMTD)的口服吸收。受到AD影响较为显著且具有临床意义的药物主要包括达沙替尼、尼洛替尼、厄洛替尼、吉非替尼、拉帕替尼、达克替尼、帕唑帕尼、博舒替尼、来那替尼、达布非尼以及舒尼替尼;受到食物影响较为显著并需要空腹服用的药物包括尼洛替尼、厄洛替尼、埃克替尼、凡德他尼、阿法替尼、依维莫司、索拉非尼、塞瑞替尼、阿来替尼、帕唑帕尼、曲美替尼、卡博替尼、达布非尼、司美替尼以及拉帕替尼;受到食物影响较为显著并需要与食物同服的药物包括瑞戈非尼、帕博西尼、来那替尼、博舒替尼以及维奈托克。应重视AD与食物对SMTD口服吸收的影响,以避免降低或增加药物的吸收,从而影响药物的有效性和安全性。 Antiacid drugs(AD)and food can influence the bioavailability of a part of small molecular targeted drugs(SMTD),in a clinically significant degree for some of them,such as dasatinib,nilotinib,erlotinib,gefitinib,lapatinib,dacomitinib,pazopanib,bosutinib,neratinib,dabrafenib and sunitinib.Drugs that have been significantly affected by food and need to be taken under a fasting condition include nilotinib,erlotinib,icotinib,vandetanib,afatinib,everolimus,sorafenib,ceritinib,alectinib,pazopanib,trametinib,cabozantinib,dabrafenib,selumetinib and lapatinib.Drugs that are significantly affected by food and need to be taken with food include regorafenib,palbociclib,neratinib,bosutinib and venetoclax.Attention should be paid to the effects of AD and food on the oral absorption of SMTD,so as to avoid reducing or increasing the absorption of drugs,and affecting the efficacy and safety of drugs.
