姜黄素水杨酰单酯增强肺癌A549细胞放射敏感性的研究

目的:研究姜黄素水杨酰单酯(FM0807)对肺癌A549细胞放射增敏性的影响,并初步探讨其作用机制。方法:将肺癌A549细胞分为空白对照组、单纯照射组、FM0807组、FM0807联合照射组。四甲基偶氮唑蓝(methyl thiazolyl tetrazolium,MTT)法测定不同浓度FM0807对肺癌A549细胞增殖的影响;克隆形成实验检测不同浓度FM0807(0,15,30和60μmol·L-1)对肺癌A549细胞放射敏感性的影响;β-半乳糖苷酶染色和流式细胞术分析FM0807(30μmol·L-1)联合射线(4 Gy)对细胞衰老率及凋亡的影响。结果:FM0807能抑制A549细胞的增殖,呈剂量-时间依赖性;随着FM0807浓度的增加,A549的放射生物学参数D0,Dq和SF2值逐渐减小,而放射增敏比SER值逐渐增加。与空白对照组、单纯照射组比较,FM0708联合照射组细胞衰老率、凋亡率均显著增加,差异具有统计学意义(P<0.01)。结论:FM0807能增强肺癌A549细胞的放射敏感性,其机制可能与促进细胞凋亡有关。

Objective:To evaluate the radiosensitization effect of curcumin salicylate monoester(FM0807)on lung cancer A549 cells and to study the primary mechanism.Methods:Cells were divided into four groups:control group,X-ray radiation group,FM0807 group and the combination group treated with X-ray plus FM0807.The effects of FM0807 at different concentrations(0,15,30,60μmol·L-1)on the cell proliferative and radiosensitivity were evaluated by MTT and colony formation assay.The effect of FM0807(30μmol·L-1)on cell senescence and apoptosis induction were detected by the Saβ-Gal staining and flow cytometry.Results:FM0807 inhibited the proliferation of A549 cells in time-and dose-dependent manners.With the increase of FM0807 concentrations,D0,Dqand SF2value of A549 cells decreased and SER value increased.Compared with control group and radiation alone group,the cell senescence and apoptosis rate significantly increased in the combination group(P<0.01).Conclusion:FM0807 can increase radiosensitivity of lung cancer cell line,A549 cells,possibly by inducing cell apoptosis.
