暴露PM2.5大鼠咽部微生态的影响 被引量:2

Effects of PM2.5 exposure on pharyngeal microecology in rats

摘要 目的探讨大气细颗粒物PM2.5对大鼠咽部微生态的影响。方法将50只清洁级健康6周龄Wistar雌性大鼠随机分为5组,每组10只,即对照组(雾化吸入生理盐水,为清洁组)、雾化吸入PM2.5浓度高低不同时间不同分为轻污短期、轻污长期、重污短期、重污长期,分别于实验前后采集大鼠咽部分泌物进行咽部微生态的检测。结果吸入PM2.5后大鼠咽部菌群中正常菌的检出率明显降低(P <0.05),而致病菌和条件致病菌检出率明显增加(P <0.05)。结论⑴在完全密闭的环境中,连续30 d给wistar大鼠雾化吸入151~300μg/m3浓度的PM2.5,可成功复制大鼠慢性咽炎的动物模型,可广泛应用于实验研究中;⑵大气细颗粒物PM2.5可以破坏大鼠咽部微生态平衡;⑶PM2.5所致的咽部微生态失衡早期主要表现为需氧菌菌群失调,晚期可发生厌氧菌菌群失调。 To investigate the effect of fine particulate matter PM2.5 on pharyngeal microecology of rats.Methods 50 healthy clean-level 6-weeks-old female Wistar rats were randomly and equally divided into 5 groups,namely the control group(atomized inhalation of physiological saline,clean group)and 4 treatment groups:light pollution short-term,light pollution long-term,heavy pollution short-term,heavy pollution long-term according to the different concentration and time of atomized inhalation PM2.5.Pharyngeal secretions of rats were collected before and after the experiment for pharyngeal micro-ecological detection.Results After PM2.5 inhalation,the detection rate of normal bacteria in pharyngeal flora of rats decreased significantly(P<0.05),while the detection rate of pathogenic bacteria and conditional pathogenic bacteria increased significantly(P<0.05).Conclusion⑴In a completely closed environment,wistar rats were atomized and inhaled PM2.5 concentration of 151~300μg/m3 for 30 consecutive days,which can successfully replicate the animal model of chronic pharyngitis in rats and can be widely used in experimental studies;⑵Atmospheric fine particulate matter PM2.5 can damage the microecological balance of the pharynx of rats;⑶The imbalance of pharyngeal microecology caused by PM2.5 was mainly characterized by the imbalance of aerobic bacteria flora in the early stage,and the imbalance of anaerobic bacteria flora in the late stage.
