不同证型变应性鼻炎复合大鼠模型鼻腔黏膜神经源性炎症特点的对比分析 被引量:5

Comparatively Study on Nasal Mucosa Neurogenic Inflammation of Different TCM Syndrome Types and Allergic Rhinitis Compound Model in Rats

摘要 目的:探讨不同证型变应性鼻炎(AR)复合大鼠模型鼻腔黏膜神经源性炎症的特点。方法:动物随机分为模型组(肺气虚AR组、脾气虚AR组、单纯AR组)与空白对照组,以相应造模方法先后建立中医证型以及证型AR复合模型。并通过证型评估、大鼠变应性鼻炎行为学评分、镜下黏膜组织病理学观察、P物质(SP)与血管活性肠肽肽(VIP)免疫组化染色以及积分光密度值测定,对各组大鼠鼻黏膜神经源性炎症特点进行对比分析。结果:中医证型造模后,肺气虚模型组大鼠呼吸频率明显升高,脾气虚模型组大鼠提质量增幅明显降低。AR复合模型建立后,各模型组大鼠行为学评分明显升高;黏膜HE染显色示肺气虚AR组炎性细胞浸润、脾气虚AR组黏膜下腺体扩张较其它组更加明显;神经肽免疫组化显示肺气虚与脾气虚AR模型组分别在SP、VIP出现高水平表达,且差异具有统计学意义(P<0.05)。结论:不同证型AR复合大鼠模型之间鼻腔黏膜神经源炎症检测指标具有一定的差异。 Objective:To investigate the characteristics of neurogenic inflammation in nasal mucosa of allergic rhinitis(AR)compound model rats with different syndrome types.Methods:The animals were randomly divided into model groups(lung-qi deficiency AR group,spleen-qi deficiency AR group,simple AR group)and blank control group.The syndrome types model of TCM and AR compound model were established by the corresponding modeling method in a time sequence.The characteristics of neurogenic inflammation in nasal mucosa of rats were assessed by TCM syndrome type evaluation.AR behavioral scores,mucosal pathological observation,immunohistochemical(IHC)staining of substance P(SP)and vasoactive intestinal peptide(VIP)and the integral optical density values in each group were observed.Results:When TCM syndrome model established,there was a significant difference in the respiratory rate increasing in the lung-qi deficiency model group and body mass increasing in the spleen-qi deficiency model group.When TCM syndrome types of AR compound model established,the AR behavioral scores of the rats in model groups were significantly increased compared with those of the blank control.HE staining of nasal mucosa showed that the inflammatory cell infiltration in the lung-qi deficiency AR group and submucosal gland expansion in spleen-qi deficiency AR group were more serious compared with those of the other groups.IHC of neuropeptides showed that there was a significant difference in SP and VIP expression in lung-qi and spleen-qi deficiency AR compound model group respectively(P<0.05).Conclusion:The compound model of AR rats with different TCM syndrome types had certain differences in the characteristics of nasal mucosal neurogenic inflammation.
