基于iTRAQ技术的黄连解毒汤对自发性高血压大鼠主动脉差异蛋白的研究 被引量:3

摘要 目的:基于iTRAQ技术分析黄连解毒汤对自发性高血压大鼠主动脉的作用蛋白,对差异蛋白进行鉴定和生物信息学分析,探讨黄连解毒汤对自发性高血压大鼠主动脉的作用机制。方法:大鼠随机分为模型组和黄连解毒汤组,分别给予相应干预后取大鼠的主动脉组织。通过iTRAQ方法分析得到差异蛋白,将相关的差异蛋白基因通过DAVID数据库搜索结合GO数据库搜索进行功能注释和富集分析,采用KEGG数据库进行信号转导通路分析,STRING数据库进行蛋白质相互作用分析。结果:共鉴定出56种差异蛋白,生物信息学分析发现,差异蛋白主要参与了26种生物学过程、20种细胞组分、15种分子途径,差异蛋白参与4条信号转导通路,差异蛋白基因的功能主要包括ATP能量代谢、氧气转运、线粒体质子转运ATP合酶,差异蛋白主要富集在氧化磷酸化信号通路上。结论:黄连解毒汤可能通过上调ATP合酶基因蛋白Q6PDU7,P19511,D3ZAF6促进线粒体氧化磷酸化功能达到保护血管的作用。 Objective:Based on iTRAQ technology to analyze the action protein of Huanglian Jiedu Decoction on the aorta of spontaneous hypertensive rats and then identify the difference protein and bioinformatics analysis and explore the action mechanism of Huanglian Jiedu Decoction on the spontaneously hypertensive rats aorta.Methods:The rats were randomly divided into model group and Huanglian Jiedu Decoction group,and the aortic tissue of the rats was taken out after the corresponding intervention.The differentially expressed proteins were obtained by iTRAQ method analysis.The relevant differentially expressed protein genes were annotated and enriched by the DAVID database combined with GO database.The signal transduction pathway was analyzed by KEGG database,and the protein interaction was analyzed by the STRING database.Results:A total of 56 differentially expressed proteins were identified.According to bioinformatics analysis,the differentially expressed proteins were mainly involved in 26 biological processes,20 cellular components and 15 molecular pathways,and the differentially expressed proteins were involved in 4 signaling pathways.The functions of differentially expressed protein genes mainly included ATP energy metabolism,oxygen transportation,mitochondrial proton transportation and ATP synthase.The differentially expressed proteins were mainly concentrated in the oxidative phosphorylation signaling pathway.Conclusion:Huanglian Jiedu Decoction may through up-regulating the ATP synthase gene protein Q6 PDU7,P19511,D3 ZAF6 promote mitochondrial oxidative phosphorylation function to protect the role of blood vessels.
