摘要
目的:探讨益气解毒化瘀小复方及其单味药对血管紧张素Ⅱ诱导的大鼠肾小球系膜细胞MMP3(GMCs),TIMP1表达及细胞外基质FN,Col-IV胶原蛋白的影响。方法:常规培养大鼠肾小球系膜细胞,RT-PCR法检测各组对大鼠肾小球系膜细胞MMP3、TIMP1的表达,双抗体ELISA夹心法检测各组对细胞外基质FN,Col-IV胶原蛋白的影响。结果:小复方组降低系膜细胞MMP3、TIMP1表达最为明显,小复方组降低细胞外基质FN,Col-IV胶原蛋白的影响最为明显,与各单味药,西药对照组比较,有显著性差异(P<0.01或P<0.05)。结论:益气解毒化瘀小复方可有效地抑制肾小球系膜细胞增殖,有效地促进系膜细胞凋亡,并优于单味药,从而达到抗肾小球硬化,保护肾脏的目的。
Objective:To investigate the effects of compound prescriptions for benefiting Qi,detoxification and removing stasis and its single herbs on the expressions of MMP3,TIMP1 and extracellular matrix FN and Col-IV induced by AngⅡin rat glomerular mesangial cells(GMCs).Methods:Rats GMCs were routinely cultured.The expressions of MMP3,TIMP1 in GMCs were detected by RT-PCR.The effect of each group on the extracellular matrix FN,Col-IV was detected by double antibodies sandwich ELISA.Results:The compound prescriptions group reduced the expressions of MMP3,TIMP1 in GMCs and the effect of extracellular matrix FN and Col-IV was mostly obvious.Compared with the single herb groups,telmisartan group had significant difference(P<0.01 or P<0.05).Conclusion:The compound prescriptions of benefiting Qi,detoxification and removing stasis can effectively inhibit the proliferation of mesangial cells,effectively promote the apoptosis of mesangial cells,and are superior to its single herbs so as to achieve anti-glomerular sclerosis and protect the kidneys.
