黄芪桂枝五物汤标准汤剂HPLC指纹图谱建立及3种成分含量测定 被引量:13

摘要 目的:制备黄芪桂枝五物汤标准汤剂,建立其高效液相指纹图谱分析方法,并测定汤中毛蕊异黄酮苷、桂皮醛、芍药苷的含量。方法:Phenomenex Luna C18色谱柱(250 mm×4.6 mm,5μm);流动相:乙腈-0.2%甲酸水溶液梯度洗脱;检测波长:278 nm;流速:1 mL/min;进样量:20μL;柱温:室温。结果:确认了10批黄芪桂枝五物汤标准汤剂有17个共有峰,相似度均大于0.90;毛蕊异黄酮苷、桂皮醛、芍药苷分别在0.58~14.5、0.053 3~19.68 8、0.1~5.0 mg/mL浓度范围内呈良好线性关系,精密度、重复性、稳定性、加样回收试验均符合含量测定要求。结论:所建立的指纹图谱分析方法稳定、可靠,可用于黄芪桂枝五物汤及其制剂的质量控制。 Objective:To prepare a standard decoction and establish an HPLC fingerprint of Huangqi Guizhi Wuwu Decoctionand determine the contents of calycosinin,cinnamaldehyde and paeoniflorin.Method:The analysis was performed on a Phenomenex Luna C18 column(250 mm×4.6 mm,5μm),gradient eluting with the mobile phase comprising of acetonitrile and water(containing 0.2%formic acid)flowing at 1.0 mL/min.And the injection volume was set at 20μL per injection detecting at 278 nm in room temperature.Results:It was confirmed that there were 17 common peaks in 10 batches of Huangqi Guizhi Wuwu Decoction,and their similarities were all greater than 0.90%.And calycosinin,cinnamaldehyde and paeoniflorin showed good linear relationship in 0.58~14.5μg/mL,0.053 3~19.688μg/mL,0.1~5.0 mg/mL,respectively.The precision,repeatability,stabilityand sample recovery tests all met the content determination requirements.Conclusion:The established method is stable and reliable and can be used for quality control of Huangqi Guizhi WuwuDecoction.
