“大黄-桃仁”药对防治粘连性肠梗阻物质基础及作用机制研究 被引量:16

摘要 目的:基于网络药理学理念探讨大黄-桃仁药对治疗粘连性肠梗阻的作用机制。方法:借助中药系统药理学分析平台(TCMSP)检索中药大黄、桃仁的化学成分和作用靶点,以口服利用度和类药性等为筛选指标,结合生物信息学手段评估大黄和桃仁的成分、靶点信息,运用Cytoscape 3.2.1构建化合物-靶点网络,最后通过DAVID进行基因本体(GO)功能富集分析和基于京都基因与基因组百科全书(KEGG)通路富集分析,研究其防治粘连性肠梗阻的作用机制。结果:从'大黄-桃仁'药对中筛选得到15个候选活性分子,作用于59个靶点,主要涉及在信号转导、抗凋亡、炎症反应参与激活免疫效应等生物学过程中的多条信号通路。结论:本研究结果初步验证大黄-桃仁药对防治粘连性肠梗阻的分子机制,为进一步深入探讨其药理学作用机制提供参考。 Objective:This study was designed to screen the targets of bioactive ingredients of Radix Et Rhizoma(RRER)and Persicae Semen(PS)and investigate the mechanism of herb pair on adhesive intestinal obstruction(AIO).Methods:The main targets of the active ingredients of PPER-PS were filtrated based on traditional Chinese medicine analysis platform system pharmacology(TCMSP).Cytoscape 3.2.1 was applied to build the ingredients-targets network of PPER-PS on adhesive intestinal obstruction.Results:Fifteen active components were predicted from the'PPER-PS'pair,which included aloe-emodin,catechin,rhein,GA119,GA120,GA121 and so on,which was applied to 59 targets,which mainly involved many signaling pathways in the biological processes,such as signal transduction,anti-apoptosis and inflammatory response involved in activating immune effect.Conclusion:This study proposes the system pharmacology method and finds out the potent combination therapeutic mechanisms of PPER-PS for AIO.This strategy will provide a new thought for the study of herb pair.
