摘要
目的:通过观察平目汤含药血清对Graves眼病眼眶脂肪细胞fas/fas L表达的影响,探讨平目汤治疗非活动期Graves眼病的可能作用机制。方法:SD大鼠给予平目汤灌胃,2次/d,连续给药3 d后制备平目汤含药血清。体外常规培养眼眶脂肪细胞,实验分为空白对照组,地塞米松组(30μg/mL),平目汤低剂量组(5%)、平目汤中剂量组(10%)、平目汤高剂量组(20%)。干预12 h后采用RT-qPCR检测各组细胞的fas/fasL mRNA表达,干预48 h后Western Blot法检测各组细胞的fas/fasL蛋白表达。结果:与空白对照组比较,平目汤中剂量组fas mRNA的表达量升高(P<0.05),平目汤各组fas L mRNA的表达量均升高(P<0.05),地塞米松组无统计学差异(P>0.05);与空白对照组相比,平目汤各组fas、fas L蛋白表达均上调,差异有统计学意义(P<0.05),地塞米松组则无统计学差异(P>0.05)。结论:平目汤治疗非活动期Graves眼病的作用可能与上调眼眶脂肪细胞fas/fas L的mRNA和蛋白表达有关。
Objective:To observe the effect of Pingmu Decoction medicated serum on the expression of fas/fas L in orbital fat cells of Graves’ophthalmopathy and explore the possible mechanism of Pingmu Decoction on inactive Graves’ophthalmopathy.Methods:SD rats were given Pingmu Decoction twice a day for 3 days to prepare the medicated serum.The orbital fat cells were cultured in vitro,and the experiment was divided into blank control group,dexamethasone group(30μg/mL),Pingmutang low dose group(5%),Pingmutang medium dose group(10%),Pingmutang high dose group(20%).The mRNA expression of fas/fas L in each group was detected by RT-qPCR after intervention for 12 hours and the protein expression of fas/fas L in each group was detected by Western blot after intervention for 48 hours.Results:Compared with the blank control group,the mRNA expression of fas in the middle dose group of Pingmu Decoction was increased(P<0.05),and the mRNA expression of fas L in Pingmu Decoction groups ware increased(P<0.05).There was no statistical difference in dexamethasone group(P>0.05).Compared with the blank control group,the protein expressions of fas and fas L in Pingmu Decoction group were up-regulated(P<0.05),and there was no significant difference in dexamethasone group(P>0.05).Conclusion:The effect of Pingmu Decoction on the treatment of inactive Graves’ophthalmopathy may be related to the up-regulation of mRNA and protein expression of fas/fas L in orbital fat cells.
