防风-乌梅配伍对哮喘模型小鼠气道重塑的影响 被引量:11

摘要 目的:观察防风-乌梅配伍对哮喘模型小鼠气道重塑的影响,并探索其机制,为进一步研究中药配伍奠定基础。方法:70只BALB/C小鼠随机分为正常对照组、哮喘模型组、激素干预组、防风组、乌梅组、防风-乌梅组、防风-乌梅合激素干预组。以卵蛋白(OVA)建立哮喘模型。造模成功后连续用药干预7 d。防风组、乌梅组、防风-乌梅组、防风-乌梅合激素干预组分别于上午9∶00给予防风(3.082 5 g·kg-1·d-1)、乌梅(3.082 5 g·kg-1·d-1)、防风-乌梅(6.165 g·kg-1·d-1)、防风-乌梅(6.165 g·kg-1·d-1)药液灌胃,其余各组给予生理盐水灌胃;激素干预组、防风-乌梅合激素干预组分别于下午3∶00给予0.02%布地奈德雾化吸入,其余各组给予生理盐水雾化吸入。观察各组小鼠肺组织病理改变,测量支气管平滑肌厚度,ELISA法测定肺组织MMP-9、TIMP-1、IL-6及IL-8的含量。结果:防风-乌梅配伍可改善哮喘模型小鼠肺组织炎症,减少支气管平滑肌厚度,降低肺组织IL-6、IL-8、MMP-9、TIMP-1的表达及MMP-9/TIMP-1的比值(P<0.05或P<0.01),以防风-乌梅合激素干预组最佳,防风-乌梅组优于防风组和乌梅组。相关性分析提示IL-6、IL-8与MMP-9、TIMP-1显著相关。结论:防风-乌梅'相须为用'配伍可改善哮喘气道重塑,其机制可能与减轻支气管平滑肌肥厚、改善气道炎症、降低肺组织MMP-9、TIMP-1的表达及MMP-9/TIMP-1的比值有关。 Objective:To observe the effect of the compatibility of Fangfeng-Wumei on airway remodeling in asthma model miceand explore its mechanismso as to lay a foundation for further study of compatibility of Chinese herbs.Method:Seventy BALB/C mice were randomly divided into normal control group,asthma model group,hormone intervention group,Fangfeng group,Wumei group,Fangfeng-Wumei group and Fangfeng-Wumei combined with hormone intervention group.Animals were established asthma model by using ovalbumin(OVA).After the success of the model,continuous medication intervention was for 7 days.At 9 a.m.,Fangfeng group,Wumei group,Fangfeng-Wumei group and Fangfeng-Wumei combined with hormone intervention group wererespectively administered with Fangfeng(3.082 5 g·kg-1·d-1),Wumei(3.082 5 g·kg-1·d-1),Fangfeng-Wumei(6.165 g·kg-1·d-1)and Fangfeng-Wumei(6.165 g·kg-1·d-1)medicine liquid.Normal control group,asthma model group and hormone intervention groupwererespectively administered with normal saline.Hormone intervention group andFangfeng-Wumei combined with hormone intervention group wererespectivelygiven 0.02%budesonide inhalation 15 min at 3 p.m.The other groups were given inhalation with normal saline.Pathological changes and bronchial smooth muscle thickness in lung tissues of each group were observed.The contents of MMP-9,TIMP-1,IL-6 and IL-8 in lung tissue were determined by ELISA.Result:The combination of Fangfeng-Wumei can improve the inflammation of lung tissue and the thickness of bronchial smooth muscle,reduce the expressions of IL-6,IL-8,MMP-9,TIMP-1 and the ratio of MMP-9/TIMP-1(P<0.05 or P<0.01).The effect of Fangfeng-Wumei combined with hormone intervention group is the best.Fangfeng-Wumei groupis superior to Fangfeng group and Wumei group.Correlation analysis showed that IL-6 and IL-8 were significantly correlated with MMP-9 and TIMP-1.Conclusion:The combination of Fangfeng-Wumei can improve airway remodeling,and its mechanism may be related to reducing bronchial smooth muscle thickness,improving airway inflammation,decreasing the expressions of MMP-9,TIMP-1 and the ratio of MMP-9/TIMP-1 in lung tissue.
