摘要
目的:探讨加减枇杷清肺饮对ICR小鼠痤疮模型NLRP3炎性小体及其下游因子caspase-1、IL-1β的影响。方法:ICR小鼠按随机数字表法分为PBS对照组、模型组、四环素组、加减枇杷清肺饮高、中、低剂量治疗组6组各6只。痤疮丙酸杆菌小鼠耳内侧皮内注射造模,造模后1 d分别给予生理盐水、四环素、加减枇杷清肺饮高、中、低剂量灌胃干预共7 d。治疗后观察皮损变化及组织病理改变,Real-time PCR和western blot分别检测NLRP3、caspase-1、IL-1β的mRNA及蛋白表达水平。结果:加减枇杷清肺饮能减轻痤疮模型的炎症,减少皮脂腺增生,降低皮损中NLRP3、caspase-1、IL-1βmRNA及蛋白表达。结论:加减枇杷清肺饮可以有效改善痤疮的炎症及皮脂腺增生,抑制NLRP3炎性小体的激活是其可能的作用机制之一。
Objective: To investigate the effect of modified Pipaqingfei Decoction on NLRP3 inflammasome in ICR mice model of acne vulgaris. Methods: ICR mice were randomly divided into 6 groups( 6 mice/group) as follows: PBS control group,model group,tetracycline group,modified Pipaqingfei Decoction high,medium and low dose treatment group. Living Propionibacterium acnes was intradermally injected into the ears of the mice.One day later,the mice were given either normal saline,tetracycline or the different dose of modified Pipaqingfei Decoction by intragastric administration for 7 days. After treatment,the changes of skin lesions and histopathological changes were observed. The levels of NLRP3,caspase-1,IL-1β gene transcription and translation were analyzed by Real-time PCR and Western blot,respectively. Results: Modified Pipaqingfei Decoction can reduce the inflammation of acne animal model,reduce sebaceous gland hyperplasia,and can reduce the expression of NLRP3, caspase-1,IL-1β gene mRNA and protein in skin lesions. Conclusion: Modified Pipaqingfei Decoction can effectively improve acne inflammation and sebaceous gland hyperplasia by inhibiting the activation of NLRP3 inflammasome.
