摘要
目的:探讨肛门洗剂对肛瘘模型大鼠创面组织修复作用及其作用机制。方法:雄性SD大鼠按随机数字表法分为正常组、模型组、肛门洗剂组和阳性对照组连续干预14 d,于实验第7、14天观察大鼠创面愈合情况,酶联免疫法(ELISA)检测各组大鼠7、14 d创面肉芽组织血管生长因子(VEGF)、低氧诱导因子-1(HIF-1)、胎盘生长因子(PLGF)、促血管生成素-Ⅰ(Ang-Ⅰ)、促血管生成素-Ⅱ(Ang-Ⅱ)、白介素-1β(IL-1β)、白介素-2(IL-2)及前列腺素E_2(PGE_2)表达,蛋白印迹法(Western blot)检测各组大鼠7 d、14 d创面肉芽组织FGF-1、FGF-2、FGF-6的表达。结果:模型组较正常组大鼠7 d、14 d创口愈较差,肉芽组织VEGF、HIF-1、PLGF、Ang-Ⅰ、Ang-Ⅱ、FGF-1、FGF-2及FGF-6表达降低,IL-1β、IL-2及PGE2表达升高;肛门洗剂组及阳性对照组较模型组大鼠7 d、14 d创口愈较好,上述血管生成相关因子、炎症因子、FGF表达均明显改善且呈时效性。结论:肛门洗剂可有效改善肛瘘模型大鼠创口愈合情况,其机制可能与降低创口炎症因子表达、提高血管生长相关因子及FGF蛋白表达有关。
Objective:To investigate the role of anal decoction in repairing wound tissue of anal fistula model rats and its mechanism.Methods:Male Sprague-Dawley rats were randomized into normal group,model group,anal lotion group and positive control group,and were continuously treated for 14 days.Wound healing of rats was observed on the 7 th and14 th days of the experiment.Enzyme-linked immunosorbent assay(ELISA)was used to detect vascular endothelial growth factor(VEGF),hypoxia-inducible factor-1(HIF-1),placenta growth factor(PLGF),and angiopoietin-I(Ang-I),angiopoietin-II(Ang-II),Interleukin-1β(IL-1β),Interleukin-2(IL-2)and Prostaglandin E2(PGE2)expression in granulation tissue of wounded rats on day 7 and day 14 of each group.Western blot was used to detect the expression of FGF-1,FGF-2 and FGF-6 in the granulation tissue of the wounded rats on the 7 th and 14 th day.Results:The wounds of model group were worse than those of normal group on the 7 th and 14 th day.The expression of VEGF,HIF-1,PLGF,Ang-I,Ang-II,FGF-1,FGF-2 and FGF-6 in granulation tissue was decreased.IL-1β,IL-2 and PGE2 expression increased;The wounds in the anal lotion group and the positive control group were better than those in the model group on the 7 th and 14 th days.The above-mentioned angiogenesis-related factors,inflammatory factors,and FGF expression were all significantly improved and showed a time-effect.Conclusion:Anal lotion can improve wound healing rate and local blood flow in the anal fistula model rats.The mechanism may be related to reducing the expression of inflammatory factor and increasing the expression of vascular growth factor,fibroblast growth factor in the anal fistula model rats.
