摘要
目的:考察知母总皂苷对佐剂型关节炎模型大鼠的干预效应,并探讨其对琥珀酸/GPR91/IL-1β通路的影响。方法:采用完全弗氏佐剂制备佐剂型关节炎动物模型,致炎第14天起用知母总皂苷干预(200 mg/kg) 28 d并每隔6 d测定足容积及足围,HE染色检查滑膜病理,试剂盒测定血清IL-1β含量及琥珀酸脱氢酶活性,高效液相仪检测琥珀酸和富马酸含量,免疫组化检测GPR91蛋白表达。结果:知母总皂苷明显抑制佐剂型关节炎大鼠足肿胀度和足围,改善滑膜增生和炎性浸润,降低IL-1β和琥珀酸含量,抑制GPR91高表达,但对琥珀酸脱氢酶活性和富马酸含量无明显改变。结论:知母总皂苷拮抗佐剂型关节炎模型大鼠足肿胀及改善滑膜损伤与其抑制琥珀酸/GPR91/IL-1β通路有关。
Objective:To investigate the therapeutical effect of anemarrhena total saponins on adjuvant arthritis model rats,and to explore its influence on succinate/GPR91/IL-1βpathway.Methods:The adjuvant arthritis rat model was established by injection of complete freund’s adjuvant.14 d after modeling,anemarrhena total saponins(200 mg/kg)was given for 28 d continuously.The swelling and circumference of rat foot were measured once every seven days and then the degree of foot swelling was calculated.The pathological change of synovial tissue was detected by HE staining.The kit was used to detect the serum IL-1βcontent and the activity of succinate dehydrogenase.The succinate and fumarate level of rat synovial tissue were determined by high performance liquid chromatography.The immunohistochemistry was applied to test the expression of GPR91 protein.Results:Anemarrhena total saponins can significantly reduce the swelling and circumference of adjuvant arthritis rat foot.And it can obviously improve the pathological characteristics of synovial tissue in adjuvant arthritis rats.Anemarrhena total saponins dropped the content of IL-1βand succinate level,inhibit GPR91 protein expression.But it had no significant effect on succinate dehydrogenase activity and fumarate content in synovial tissue.Conclusion:Anemarrhena total saponins can antagonize foot swelling and improve synovial damage in adjuvant arthritis model rats,which related to the inhibition of succinate/GPR91/IL-1βpathway.
